Bovine tuberculosis (BTB) is a zoonosis caused by the bacterium Mycobacterium bovis, which induces the development of nodular and granulomatous lesions in various animal tissues. The recognition of these suggestive gross lesions during postmortem sanitary inspection in slaughterhouses provides a presumptive diagnosis, which requires the use of complementary tests to confirm the disease. This study aimed to verify the occurrence of BTB in cattle slaughtered in slaughterhouses in the state of Ceará, Brazil, using bacteriological and molecular methods. To this end, suggestive lesions were analyzed on carcasses condemned by the “Serviço de Inspeção Estadual” (SIE). The samples were submitted to microbiological analysis using culture media and specific staining followed by spoligotyping molecular technique for identification and genotyping of the mycobacteria. Occurrence of lesions suggestive of BTB was verified in bovine carcasses (0.071%) from different municipalities of the state. These lesions were located mainly in the lung (95.12%), lymph nodes (58.53%), and liver (36.58%). Microbiological culture showed bacterial isolation (17.94%), with the growth of colonies showing morphological and tannic characteristics belonging to genus Mycobacterium spp. Genetic polymorphism analysis identified M. bovis in all isolates, which were discriminated into six spoligotypes (SB0121, SB0295, SB1064, SB0120, SB0870, and SB0852). These profiles have been described in Brazil and several areas of the world, except for profiles SB1064 and SB0852, which were described in the country for the first time. The results show that the association of the diagnostic methods used was the basis for the first study on identification of mycobacteria found in the state, which may provide a database for the epidemiological study of BTB in the state of Ceará.

RESUMO: Este estudo teve como objetivo avaliar lesões sugestivas de tuberculose em búfalos abatidos em matadouros oficiais no Estado do Amapá, Brasil, a fim de confirmar o diagnóstico de tuberculose por avaliação histopatológica e molecular. As amostras de tecido de 20 búfalos que apresentavam lesões sugestivas de tuberculose, dos municípios de Macapá e Santana, foram coletadas. As amostras foram divididas em duas partes: uma delas foi fixada em formalina a 10% tamponada e rotineiramente processadas para avaliação histopatológica, coradas pela hematoxilina-eosina e Ziehl-Neelsen; e o outra parte foi usado para Nested-PCR para o complexo de Mycobacterium tuberculosis (CMT) e para Mycobacterium bovis. As lesões macroscópicas sugestivas de tuberculose foram observadas nos pulmões, linfonodos brônquicos, mediastínicos, retrofaríngeos e submandibulares, fígado e pleura. Histopatologicamente, todas as amostras apresentaram lesões sugestivas de tuberculose, caracterizadas por granulomas compostos por grande quantidade de infiltração de células epitelióides, células de Langerhans e linfócitos, margeando um centro necrótico, calcificado ou não, rodeado por cápsula de tecido conjuntivo fibroso. Bacilos álcool-ácido resistentes foram observados nos tecidos de 3/20 (15%) búfalos. Com relação à detecção molecular, 13/20 (65%) bubalinos apresentaram amostras de tecidos positivos: 6 foram positivos nas Nested-PCRs para CMT e M. bovis, um foi positivo apenas na Nested-PCR para CMT, e 6 foram positivos apenas na Nested-PCR para M. bovis. Os resultados deste estudo demonstram a importância de diagnosticar a tuberculose em búfalos na região e apontam para a necessidade de implementar medidas eficazes para controlar e erradicar a enfermidade.
Uma doença preocupante tanto para os animais quanto para o homem, a tuberculose está amplamente difundida nos rebanhos bovinos ao redor de todo o país, gerando grandes prejuízos econômicos para os produtores e riscos sanitários para o ser humano. Apresentar a prevalência da tuberculose bovina em animais abatidos em frigoríficos rondonienses possuintes do Serviço de Inspeção Federal (SIF) foi o objetivo do presente trabalho. Demonstrando de modo retrospectivo as taxas de prevalência da tuberculose em bovinos abatidos em Rondônia nos anos de 2011 à 2017 e, por final, estipular uma média geral da prevalência da patologia no rebanho abatido no estado através dos anos aqui ponderados. Para a realização das análises estatísticas todos os dados citados foram fornecidos pela Agência de Defesa Sanitária Agrosilvopastoril do Estado de Rondônia (IDARON), no mês outubro de 2018. Foi disponibilizado: quantidade de animais abatidos em frigoríficos possuidores do SIF, animais positivos para tuberculose e população total de bovinos em Rondônia, sendo todas as informações do período de 2011 à 2017. Tais informações foram compiladas, calculadas e organizadas em tabelas. Para se chegar ao resultado da prevalência, o cálculo utilizado foi o de divisão () da quantidade total de animais abatidos pelos animais positivos confirmados por testes laboratoriais. Assim, a prevalência da tuberculose bovina nestes frigoríficos, em porcentagem (%), foi de: 0,00085 / 2011; 0,00033 / 2012; 0,00101 / 2013; 0,00141 / 2014; 0,00103 / 2015; 0,00114 / 2016; 0,00059 / 2017. Podendo-se afirmar que a prevalência da tuberculose em frigoríficos rondonienses que possuem o SIF se encontra demasiadamente baixa.
