Withania somnifera (Ashawagandha) is very revered herb of the Indian Ayurvedic system of medicine as a Rasayana (tonic). It is used for various kinds of disease processes and specially as a nervine tonic. Considering these facts many scientific studies were carried out and its adaptogenic / anti-stress activities were studied in detail. In experimental models it increases the stamina of rats during swimming endurance test and prevented adrenal gland changes of ascorbic acid and cortisol content produce by swimming stress. Pretreatment with Withania somnifera (WS) showed significance protection against stress induced gastric ulcers. WS have anti-tumor effect on Chinese Hamster Ovary (CHO) cell carcinoma. It was also found effective against urethane induced lung-adenoma in mice. In some cases of uterine fibroids, dermatosarcoma, long term treatment with WS controlled the condition. It has a Cognition Promoting Effect and was useful in children with memory deficit and in old age people loss of memory. It was also found useful in neurodegenerative diseases such as Parkinson's, Huntington's and Alzeimer's diseases. It has GABA mimetic effect and was shown to promote formation of dendrites. It has anxiolytic effect and improves energy levels and mitochondrial health. It is an anti-inflammatory and antiarthritic agent and was found useful in clinical cases of Rheumatoid and Osteoarthritis. Large scale studies are needed to prove its clinical efficacy in stress related disorders, neuronal disorders and cancers.

Stress is a state of mental or emotional strain resulting from demanding circumstances. It's a universal experience, which has an important defensive function towards harmful psychological stimuli. Owing to this there are various pharmacological and non-pharmacological treatments with fatal adverse reaction. The need of hour is to have a cost-effective adaptogenic drug. Adaptogens are compounds that increase the ability of an organism to adapt to environmental factors and avoid damage. Ayurveda explains these qualities under concepts of vyadhikshamatva (immunity). Ashwagandha is considered to be the best adaptogenic drug among shankhapushpi (Clitoria ternatea), yashtimadhu (Glycyrrhiza glabra), brahmi (Bacopa monnieri), guduchi (Tinospora cordifolia), mandookaparni (Centella asiatica), etc medhya dravya (cognitive enhancers). This study reviews the Ayurvedic and modern literature related to stress and its management as adaptogens through Ayurveda. Ayurveda is the traditional system of medicine of India and contains a very scientific description of stress and general manovikaras (psychiatric illness). Ayurveda has been a great potential in the field of rasayana (rejuvenation) as there are lots of single herbs and preparations which possess adaptogenic properties and strengthens the physiological adaptation.

“…Back in 1965, Malhotra et al found that, the oral LD 50 of a 2 % suspension of ashwagandholine (total alkaloids from the roots of WS) prepared in 10 % propylene glycol using two percent gum acacia as suspending agent was 465 mg/kg (332-651 mg/kg) in rats and 432 mg/kg (299-626 mg/kg) in mice. Moreover, the acute oral LD 50 of alcohol extract from defatted WS seeds dissolved in normal saline was 1750 ± 41 mg/kg in albino mice (Singh, et al 1982). Toxicity studies on WS are very limited. The purpose of the present work was to study the acute and sub-chronic toxicity of WS extracts in rats and to determine the LD 50 value, which in turn helps in determining the dose range for the plant extract to be used with no harmful or lethal effects on the animal. Rats in acute study were IP injected with the alcoholic extract of WS (the aerial parts) at doses of (0), 150, 300, 600 and 1200 mg/kg bwt. In subchronic toxicity study WS extract was administered IP at a dose of 10%, 20% and 40% of the obtained LD 50 (52 mg/kg bwt, 104 mg/kg bwt and 208 mg/kg bwt respectively) for 60 days. Acute toxicity revealed an IP LD 50 of 522 mg/kg wt. Significant changes in body weight, hematological, biochemical and histopathological changes were recorded in 20% and 40% LD 50 groups with 15 -40 % mortalities respectively, by the end of the experiments. Based on this study, WS showed toxicity at doses higher than 10% LD 50 when given IP and it was suggested to use ≤ 10% LD 50 (IP) doses of WS extracts, than the tested here to start with in setup protection or treatment studies.Correspondence to: Khaled M.Ashry : ashrykh@yahoo.com
