Phasercrystallographic software

Abstract: Phaser is a program for phasing macromolecular crystal structures by both molecular replacement and experimental phasing methods. The novel phasing algorithms implemented in Phaser have been developed using maximum likelihood and multivariate statistics. For molecular replacement, the new algorithms have proved to be significantly better than traditional methods in discriminating correct solutions from noise, and for single-wavelength anomalous dispersion experimental phasing, the new algorithms, which account… Show more

“…The McjD‐ADP‐VO 4 structure was determined by molecular replacement in Phaser (McCoy et al , 2007) using our previously published McjD structure (PDB ID: 4PL0; Choudhury et al , 2014) as search model. Initial refinement to 3.4 Å was carried out in REFMAC5 (Murshudov et al , 1997) and at later stages in Buster (Blanc et al , 2004).…”

“…The datasets were integrated and scaled using the HKL-2000 software [58]. The initial phases were calculated by the molecular replacement method using the software Phaser [59]. The coordinate of the anti-OspA Fab fragment (PDB ID: 1FJ1) was used as a search model.…”

“…Refinement was attempted at higher resolution, but this resulted in significantly higher R‐factors and noisy maps. The Se‐SAD structure was used as a model for molecular replacement with PHASER 61, the output of which was again refined using COOT and REFMAC5. Geometric restraints were relaxed somewhat relative to those recommended by Engh and Huber 62 based on recommendations by Jaskolski et al .…”
