In vitro antimicrobial effect of bacteriophages on human dentin infected with Enterococcus faecalis ATCC 29212

Abstract: This study assessed the effect of bacteriophages on the viability of Enterococcus faecalis. Human dental roots were inoculated with a suspension of E. faecalis at three different multiplicities of infection - 0.1, 1.0 and 10.0. The phage lysate was able to significantly inhibit bacteria growth when incubated at the multiplicities of infection of 1.0, 10.0 and 0.1. The dental roots were also inoculated with bacteria for 6 days to allow bacterial penetration into the teeth tubules. Addition of the phage lysate t… Show more

“…Although several bacteriophages have been isolated for a number of oral bacteria (8,10,12,13,23,33), there have been no reports on the isolation of a phage for fusobacteria until now. F. nucleatum, together with a bacterial consortium, generates the well-known dental plaque, a biofilm conformation that eventually leads to the onset of periodontal disease.…”

“…There are many examples of the use of bacteriophages at a clinical (14,32) and commercial level (20). Specifically in the dentistry area, several bacteriophages that infect diverse oral bacteria have been isolated from saliva and dental plaque (12,13,23,37). Although F. nucleatum is an important periodontal pathogen, reports of bacteriophages for this microorganism do not exist.…”

“…Despite the significance of E. faecalis in root canal infections, very few attempts were made to treat E. faecalis infection using phage therapy (31,32). In the present study, an E. faecalis-infecting phage was isolated from sewage and characterized genetically by sequencing and functionally by evaluating its efficacy against E. faecalis planktonic and biofilm cultures in vitro.…”

“…This may have affected the ability to recover viable bacteria post-treatment for phage efficacy quantification. Studies that had used successfully longer or multiple treatment times (129,130,132) may have used a variant phage. The 24h phage treatment time used in this study allowed for comparison with other research that also used this interaction period (37,39,40,42,132,149).…”
