Hyperspectral phasor analysis enables multiplexed 5D in vivo imaging

Abstract: Time-lapse imaging of multiple labels is challenging for biological imaging as noise, photobleaching and phototoxicity compromise signal quality, while throughput can be limited by processing time. Here, we report software called Hyper-Spectral Phasors (HySP) for denoising and unmixing multiple spectrally overlapping fluorophores in a low signal-to-noise regime with fast analysis. We show that HySP enables unmixing of seven signals in time-lapse imaging of living zebrafish embryos.

“…Future experiments using novel faster imaging techniques, like lattice light sheet imaging, that image fast endosomal dynamics along with technologies that can image multiple components simultaneously, such as hyper spectral imaging, might answer the plethora of questions that this fantastic biological system poses. It will be imperative to continue mapping the various lipids and protein machineries and their effectors at the endosome in the context of cargoes, and to measure their spatiotemporal distribution and dynamics by using modern technologies …”

“…Here we address this task for imaging studies of developing tissues, where patterns of cell fates are established by complex regulatory networks [6][7][8]. Advances in live imaging continue to provide new insights into the dynamics of individual components in these networks, but imaging more than three reporters at the same time is still challenging and limited to model genetic organisms [9,10]. Furthermore, in the absence of reliable live reporters, dynamics of some state variables can only be inferred from fixed tissues.…”
