Abstract: Gelatin–hyaluronic acid (Gel–HA) hybrid hydrogels have been proposed as matrices for tissue engineering because of their ability to mimic the architecture of the extracellular matrix. Our aim was to explore whether tyramine conjugates of Gel and HA, producing injectable hydrogels, are able to induce a particular phenotype of encapsulated human mesenchymal stem cells without the need for growth factors. While pure Gel allowed good cell adhesion without remarkable differentiation and pure HA triggered chondrogen… Show more

“…Jiang et al demonstrated superior chondrogenic differentiation of MSC embedded in Col compared to MSC only in a rat model [50]. On the other hand, HA has been combined with Col 1 or gelatin before with similar outcomes that chondrogenic differentiation is promoted in the composite compared to HA or Col only [51][52][53]. One limitation of the present technique is the shear-induce alignment, limiting the degrees of spatial freedom in fibril arrangement.…”

“…For instance, Pfeifer et al evidenced that human MSCs embedded in HA-GEL hydrogels synthetized cartilage molecules such as collagen type II, GAGs, TGFbeta and melanoma inhibitory activity protein (MIA) [92]. Similarly, some studies have shown that chondrogenesis is improved in the absence of growth factors such as TGF-β3, as was the case with the mixture of 70% HA -30% GEL reported by Moulisová et al, [85], the collagen -HA scaffolds developed by Murphy et al, [330], and the decellularized cartilage matrices investigated by Burnsed et al, [331]. Other feature to highlight from the HA-GEL hydrogels is the ability to precisely control physiochemical and mechanical properties such as shear modulus, stiffness, water permeability and swelling and crosslinking degree [85], [332]- [334].…”

“…According to the aforementioned, a 3D structure composed by hyaluronic acid provides greater hydration and a better diffusion of nutrients. Moreover, the content of this polysaccharide not only increases the mechanical properties of the construct such as greater support to compression, tension and shear loads, but also enhances the physicochemical properties such as swelling degree, viscoelasticity and permeability [85]. Additionally, if the 3D structure contains gelatin, it will generate an ideal environment in which the cells can adhere to the ECM.…”
