HLA-G-mediated NK cell senescence promotes vascular remodeling: implications for reproduction

Abstract: The uterus in early pregnancy is a non-lymphoid organ that is enriched in natural killer (NK) cells. Studies to address the role of these abundant human NK cells at the maternal/fetal interface have focused on their response to the major histocompatibility complex (MHC) molecules on fetal trophoblast cells that they contact. The interaction of maternal NK cell receptors belonging to the killer cell immunoglobulin-like receptor (KIR) family with trophoblast MHC class I molecules in pregnancy can regulate NK cel… Show more

“…The expression of HLA-G, a proposed ligand for KIR2DL4 (47), at the maternal-fetal interface has led to the suggestion that KIR2DL4 is involved in the maintenance of pregnancy. It is postulated that this interaction leads to the activation of NK cells and the secretion of factors that promote vascular remodeling in the uterus (48,49). To this end, some groups have reported an interaction with HLA-G, whereas other investigations suggest that it does not (23,24).…”

“…HLA-E can combine with another type of inhibitory receptor on NK cells, NKG2A, which limits the killing activity of NK cells. HLA-G has a function similar to that of HLA-E. Further study found that HLA-G can also induce tolerance of DCs and induce NK cells to secrete cytokines and angiogenesis factors, thus showing polyfunctionality (LeMaoult et al, 2004;van der Meer et al, 2004;Li et al, 2009;Rajagopalan, 2014). At the surface of the second interface, the syncytial trophoblast cells do not express MHC molecules; therefore, they cannot induce an antigen specific response from maternal T cells (Moffett-King, 2002).…”

“…In particular, most of previous studies on NK cells have focused on mouse splenic and human peripheral blood NK cells, which are circulating and referred to as conventional NK (cNK) cells (Sojka et al, 2014b). However, NK cells have recently been attracting attention for their heterogeneous subsets based on tissue distribution (Rajagopalan, 2014;Shi et al, 2011).…”
