The adrenal gland is a vital endocrine gland that secretes many important hormones in everyday bird life. The adrenal gland of the Japanese quail is grossly located ventromedially the corresponding kidney and has a creamy to yellow color. The quail gland is surrounded by a capsule and contains some ganglionic cells, and the capsule is characterized by the presence of chromaffin cells. The adrenal gland is subdivided into three concentric zones: subcapsular, peripheral, and central. The parenchyma consists of interrenal tissue, chromaffin islets, and blood sinusoids. The interrenal cells contain lipid droplets, are arranged in cords, and rest on the basement membrane. Chromaffin cells are categorized as two types: epinephrine (E) and norepinephrine (NE) cells. These cells contain the granules, and are characterized by the presence of lipid droplets. In this study, the interrenal tissue was found to have a higher proportion of chromaffin cells in quail as compared with other birds, which is attributed to the fact that the Japanese quail is a migratory bird. Therefore, the present investigation aims to provide a detailed study on the adrenal gland in the Japanese quail to help physiologists understand the gland's function and the pathologist to determine the implications for the differential diagnosis of adrenal gland tumors. Highlights The adrenal gland is subdivided into three concentric zones: subcapsular, peripheral, and central. The interrenal cells contain lipid droplets, are arranged in cords. Chromaffin cells are categorized as two types: epinephrine (E) and norepinephrine (NE) cells. These cells contain the granules, and are characterized by the presence of lipid droplets. The adrenal gland responds to heat stress by epinephrine and glucocorticoid release to alleviate the adverse effects. This study investigated the effect of acute heat stress on the protein profile and histone modification in the adrenal gland of layer-type country chickens. A total of 192 roosters were subject to acute heat stress and thereafter classified into a resistant or susceptible group according to body temperature change. The iTRAQ analysis identified 80 differentially expressed proteins, in which the resistant group had a higher level of somatostatin and hydroxy-δ-5-steroid dehydrogenase but a lower parathymosin expression in accordance with the change of serum glucocorticoid levels. Histone modification analysis identified 115 histone markers. The susceptible group had a higher level of tri-methylation of histone H3 lysine 27 (H3K27me3) and showed a positive crosstalk with K36me and K37me in the H3 tails. The differential changes of body temperature projected in physiological regulation at the hypothalamus–pituitary–adrenal axis suggest the genetic heterogeneity in basic metabolic rate and efficiency for heat dissipation to acclimate to thermal stress and maintain body temperature homeostasis. The alteration of adrenal H3K27me3 level was associated with the endocrine function of adrenal gland and may contribute to the thermotolerance of chickens.

The analysis of scientific literature shows that the study of adrenal morphology in humans and animals is an urgent problem of biology and medicine from ancient times to the present day. In the historical aspect, we can distinguish three stages of studying the adrenal gland that differ in the directions of research. The first stage corresponds to the research of scientists of the XVI–XVIII centuries (Bartolomeo Eustachio, Andreas Vesalius, Gabriele Folloppio, Girolamo Fabrici, Andriaan Spieghel, Johann Vesling, Giulia Casseria, Caspar Bauhin, Johann Grafenberg, Caspar Bartholin, Thomas Bartholin, Giulio Casserio, Antonio Molinetti, Jean Riolan, Thomas Wharton, Giovanni Lancisi, Jakob Winslow, Antony Valsalva, Albrecht Haller, Johann Meckel, Jean Senac, Armand Cassan). Their works is devoted to the study of topography, macroscopic structure and function of the adrenal gland. Studies of morphological scientists of the second stage (late XVIII – mid XX century) correspond to the study of the structure of the adrenal gland at the microscopic level. Scientists like Albert Kelliker, Johann Ecker, Thomas Addison, Gabriel Colin, Alfred Kohn and Jay Arnold used histological and histochemical research methods. Edward Schafer, George Oliver, Vladislav Szymonowicz, John Abel, Jokiti Takmine, Welter Cannon, Edward Kendall to the study of the features of adrenal hormone secretion. Scientific works of the mid-twentieth and early twenty-first centuries can be combined in the third stage of research on the morphology of the adrenal gland. It is devoted to solving issues related to the formation of the adrenal gland in the process of filogenesis and ontogenesis, the features of the structure and function of the organ in normal and pathological conditions. A significant contribution to the study of the morphology of the adrenal gland of animals at the third stage was made by such scientists as Ziyade A. M., Dardykina O. N., Harina V. V., Atagimov M.
