High-Throughput Glycomic Methods

Abstract: Glycomics aims to identify the structure and function of the glycome, the complete set of oligosaccharides (glycans), produced in a given cell or organism, as well as to identify genes and other factors that govern glycosylation. This challenging endeavor requires highly robust, sensitive, and potentially automatable analytical technologies for the analysis of hundreds or thousands of glycomes in a timely manner (termed high-throughput glycomics). This review provides a historic overview as well as highlights … Show more

“…In this technique, lectins are immobilized on a solid surface, and binding of target glycoproteins can be detected either directly through their prior labeling with fluorescent reagents, or indirectly by overlaying a fluorescently labeled relevant antibody raised against the target glycoprotein (or via biotinylated antibody and fluorescently labeled streptavidin). The microarray is scanned, followed by the interpretation of the signals [ 71 , 72 ], and structural information about the glycome is obtained using the known glycan-binding specificities of the lectins [ 71 ]. The utilization of multiple lectins in the microarray allows the studying of multiple lectin–glycan interactions in a single experiment and holds the promise of enabling glycomic profiling of cancers in a fast and efficient manner [ 73 ].…”

“…Another key point regarding data collection is the current limited availability of quantitative data that would allow more accurate profiling. At present, immunoglobulin profiling is by far the most advanced in comparison to other glycoconjugates. , Other prospects can be expected to expand from improved techniques in glycan and tissue imaging. , …”

“…With regard to the role of SARS-CoV-2 glycosylation in viral replication, infectivity and immune response, glycosylation has major potential implications for therapeutic and vaccination strategies as well as serological testing [23]. Historically, the role of glycosylation in the dichotomous immune system has always been the focus of research [24][25][26][27][28][29].…”
