High-resolution DNA Fiber-FISH for genomic DNA mapping and colour bar-coding of large genes

Abstract: We have applied two-colour fluorescence in situ hybridization (FISH) to DNA fibers and combined it with digital imaging microscopy for the mapping of large cosmid contigs. The technique was validated using a set of unique plasmids and a cosmid contig both originating from the thyroglobulin (Tg) gene and previously mapped by restriction analysis. The resolution proved to be close to the theoretical lower limit of approximately 1 kb, ranging > or = 400 kb. Subsequently a 400 kb cosmid contig derived from a DMD-Y… Show more

“…So, the Watson-Crick standard method has been used for several studies (PARRA and WINDLE 1993;Florijn et al 1995;HEISKANEN et al 1995;S J~B E R G et al 1997) to estimate the size of clones and the physical distances between them. The estimations from fiber-FISH mapping have been found to be in good agreement with the one from restriction mapping by PFGE analysis HEISKANEN et al 1995;SJOBERG et al 1997).…”

“…This technique is based on the hybridization of probes to unfixed linearized DNA fibers on glass slide, and has been used to establish the order, orientation and size of several probes (PARRA and WINDLE 1993;HOULEAS et al 1994;HAAF and WARD 1994;FLORIJN et al 1995;KLOCKARS et al 1996;SJOBERG et al 1997;SHIELS et al 1997), to rapidly identify aberrations and rearrangements of genes (HEISKANEN et al 1995) and also to directly visualize the DNA replication (ROSENBERG et al 1995) and exon mapping (FLORIJN et a]. 1996).…”

“…There are diferent applications which used FISH-based methods like reverse-FISH [27], ibre-FISH [28,29], M-FISH (multicolor FISH) [30], SKY (spectral karyotyping FISH) [31], low-FISH [32], Q-FISH (quantitative FISH) [33], COBRA-FISH (combined binary ratio labelling FISH) [34], cenM-FISH (centromere-speciic M-FISH) [35], podFISH (parental origin determination FISH) [36] and heterochromatin-M-FISH [37]. The most advanced FISH-based approaches included COBRA-FISH, M-FISH and SKY.…”
