The health, economy, and quality of life all over the world are greatly affected by bacterial infections and viral outbreaks. Bacterial cells and viruses, such as influenza, can spread through contaminated surfaces and fomites. Therefore, a possible way to fight these pathogens is to utilize antibacterial and antiviral coatings, which reduce their numbers on contaminated surfaces. Here, we present a novel short peptide that can self-assemble, adhere to various surfaces, and bind different metal ions such as copper, which provides the surface with antibacterial and antiviral properties. For these functions, the peptide incorporates the amino acid 3,4-dihydroxyphenylalanine (DOPA), which provides the peptide with adhesive capabilities; a diphenylalanine motif that induces the self-assembly of the peptide; the metal-binding hexahistidine sequence. Our results demonstrate that the coating, which releases monovalent cuprous ions and hydrogen peroxide, provides the surfaces with significant antibacterial and antiviral properties. Additionally, the coating remains transparent, which is favorable for many objects and especially for display screens.

Because of their high specificity and low side effects, protein drugs possess a substantial global market. However, the low bioavailability of protein is still a major obstacle to their expanded applications, which is expected to be answered with proper protein formulations. Taking corneal neovascularization (CNV) as an example, we demonstrated a co-assembled system of hexa-histidine and Ava (Avastin) with metal ions (HmA@Ava) could cross the cornea, the most important bio-barrier during the treatment of most diseases of the anterior segment in clinics. We found that the nanosized HmA@Ava efficiently encapsulated Ava with impressive loading capacity without destroying the bioactivity of Ava and assisted Ava penetration through the corneal barriers to effectively inhibit CNV development in an alkali burn rat model with sustained and pH-dependent Ava release. Our results suggested that the co-assembled strategy of protein and HmA is a proper formulation to protein drugs, with promising penetration ability to deliver protein across bio-barriers, which could open a path for topical administration of protein drugs for treatment of various ocular diseases and hold enormous potential for delivery of therapeutic proteins not only for ocular diseases but also for other diseases that require protein treatment.

Diabetes has been listed as one of the three major diseases that endanger human health. Accurately injecting insulin (Ins) depending on the level of blood glucose (LBG) is the standard treatment, especially controlling LBG in the long‐term by a single injection. Herein, the pH‐responsive hexa‐histidine metal assembly (HmA) encapsulated with enzymes (GOx and CAT) and Ins (HmA@GCI) is engineered as the vehicle for glucose‐mediated insulin delivery. HmA not only shows high proteins loading efficiency, but also well retained proteins activity and protect proteins from protease damage. Within HmA, the biocatalytic activities of enzymes and the efficiency of the cascade reaction between GOx and CAT are enhanced, leading to a super response to the change of LBG with insulin release and efficient clearance of harmful byproducts of GOx (H2O2). In the treatment of diabetic mice, HmA@GCI reduces LBG to normal in half an hour and maintains for more than 5 days by a single subcutaneous injection, and nearly 24 days with four consecutive injections. During the test period, no symptoms of hypoglycemia and toxicity to tissues and organs are observed. These results indicate that HmA@GCI is a safe and long‐acting hypoglycemic agent with prospective clinical application.
