We genotyped 23 Y-STR loci (DYS576, DYS389I, DYS448, DYS389II, DYS19, DYS391, DYS481, DYS549, DYS533, DYS438, DYS437, DYS570, DYS635, DYS390, DYS439, DYS392, DYS643, DYS393, DYS458, DYS385a/b, DYS456, and GATA-H4) in a sample of 109 unrelated male Chinese people residing in Minnan area and compared the results with those from our previous study on 12 Y-STR. The haplotype diversity and the discrimination capacity of the 23 Y-STR reached 0.9903 and 0.9725, respectively, and the genetic diversity for each locus ranged from 0.321 (DYS391) to 0.955 (DYS385). Besides, we observed a strong correlation between the number of Y-STR markers and the substantial improvement of forensic parameters used to discriminate between individuals. The results indicated that these highly polymorphic Y-STR markers were useful for human identification in forensic cases and paternity tests within the Minnan Han Chinese population.

Population study of thrombophilic markers and pharmacogenetic markers of warfarin prevalence in Bosnia and Herzegovina

Aim To investigate the prevalence of common genetic variants that can serve as markers of thrombophilia and warfarin pharmacogenetics in Bosnia and Herzegovina. Methods The study was performed between August and October 2017 on 130 healthy unrelated adult volunteers from Bosnian-Herzegovinian population sample. The prevalence of the following genetic variants was determined: F5 c.1601G>A (factor V Leiden), F2 c.*97G>A (factor II or prothrombin mutation), F13A1 (factor XIII) c.103G>T, MTHFR (methylenetetrahydrofolate reductase) c.665C>T and c.1286A>C, as well as PAI-1 (plasminogen activator inhibitor 1) c.-816A>G and c.-844G>A as markers of thrombophilia risk, and *2 and *3 alleles of CYP2C9 (cytochrome P450 2C9) and five variants of VKORC1 (vitamin K epoxide reductase complex subunit 1) as markers of warfarin pharmacogenetics. DNA was isolated from buccal swabs using salting out method, while genotyping was performed using matrix-assisted laser desorption/ionization-time-of-flight mass spectrometry. Results Minor allele frequencies for two main thrombophilia risk factors, F5 c.1601G>A and F2 c.*97G>A were 0.023 and 0.008, respectively. Combined data for the markers of warfarin pharmacogenetics imply that 57.4% study participants can be expected to metabolize warfarin at an extensive, 40.3% at intermediate, and 2.3% at a poor rate. Conclusion This study reports the first extensive population genetic data for thrombophilia and warfarin pharmacogenetic markers in Bosnia and Herzegovina. Allele frequencies of genetic variants are within the general average for European populations, and their presence implies the necessity of introduction of personalized medicine in warfarin-mediated antithrombotic therapy.

Orofacial diseases of multidiscipline nature from carious lesions to tumors are significant diseases attack a large number of individuals around the world. However, In the recent years, bioinformatics methods have been reported to analyses several loci of simultaneously. Moreover, Analysis of molecular variance (AMOVA) (23) was performed to measure the ge netic distance between the Iraqi popu lation sample and reference data from populations. In this study, we analyzed the genetic polymorphisms of 23 Y-STR loci from PowerPlex® Y23 system in 313 unrelated healthy male individuals from Iraqi populations, and observed 283 different haplotypes including 269 unique haplotypes and 16 duplicate haplotypes. in which 283 different haplotypes was identified classified into 264 (93, 23%) as a unique Haplotype and 19 (6, 77%) as replicated haplotype among individuals. The highest haplotype were found in sample H45, H85, H135,H213 and H241 at frequency 0.010 ( haplotype replicated 3 times ), While 264 haplotype with 0.0033 frequency and haplotype diversity was 0.996 with a discrimination capacity 0.93 respectively. The gene diversity values ranged from 0.3942 at DYS438 to 0.9607 at DYS385a/b. Such results indicated that the 23 Y-STR loci were highly polymorphic in Bagdad population and played crucial roles in forensic application as well as population genetics. For the first time, we reported the genetic diversity of male lineages in Bagdad population at a high-resolution level of 23 Y-STR set and consequently contributed to familial searching, Odontologic science application tracking, and anthropology analysis of Bagdad population.
