Halovivax asiaticus gen. nov., sp. nov., a novel extremely halophilic archaeon isolated from Inner Mongolia, China Strain EJ-46 T , a novel pleomorphic, aerobic, extremely halophilic member of the Archaea was isolated from sediment of the saline Lake Ejinor, in Inner Mongolia, China. This organism was neutrophilic and required at least 15 % (2?5 M) NaCl for growth. MgCl 2 was not required. The isolate was able to grow at pH 6?0-9?0. Optimum growth occurred in media containing 20 % (3?4 M) NaCl at pH 7?0-7?5. Polar lipid analysis revealed the presence of phosphatidylglycerol and phosphatidylglycerol phosphate methyl ester, derived from both C 20 C 20 and C 20 C 25 glycerol diethers. Four glycolipids were detected, one of which may be novel. The DNA G+C content was 60?3 mol%. 16S rRNA gene analysis revealed that strain EJ-46 T was a member of the phylogenetic group defined by the family Halobacteriaceae, and the highest 16S rRNA gene similarity values of 94?9 and 94?8 % were obtained with the haloalkaliphilic species of the genus Natronococcus, Natronococcus occultus and Natronococcus amylolyticus, respectively. Based on the phenotypic, genotypic and phylogenetic analyses, it is proposed that the novel isolate should be classified as representing a new genus and species, for which the name Halovivax asiaticus gen. nov., sp. nov. is proposed. The type strain is EJ-46 T (=CGMCC 1.4248 T =CECT 7098 T ).The extremely halophilic, aerobic members of the Archaea are classified within the family Halobacteriaceae, order Halobacteriales, with 20 currently recognized genera: Halalkalicoccus Members of the Halobacteriaceae are usually pink-to redpigmented because of the presence of carotenoids (Kamekura & Dyall-Smith, 1995;Hezayen et al., 2001;Grant et al., 2001). They have been isolated from various hypersaline environments, such as saline lakes (e.g. Franzmann et al., 1994;Norton et al., 1993). The haloarchaea are the most halophilic organisms known and are dominant in hypersaline environments in which the salt concentration exceeds 25 % (w/v) (Rodriguez-Valera et al., 1981). They often appear at such high density that they impart a typical red colour to the hypersaline brines.Hypersaline environments are commonly found in China. In addition to many coastal salterns, a number of salt lakes, soda lakes and salt-rich deserts are located in various geographical areas of China, e.g. Xinjiang, Inner Mongolia and Tibet Autonomous Regions. A number of new members of the Halobacteriaceae have been isolated from these saline environments (Xu et al., 1999(Xu et al., , 2001Xin et al., 2000Xin et al., , 2001Fan et al., 2004;Feng et al., 2004Feng et al., , 2005. A Gram-negative, aerobic, neutrophilic and extremely halophilic archaeon (strain AJ201 T ), isolated from Ayakekum salt lake on the Qinghai-Tibet Plateau, was investigated by a polyphasic approach. The DNA G+C content of strain AJ201 T was 65.7 mol%. The major polar lipid profile and phylogenetic analysis based on 16S rRNA gene sequences supported the allocation of the strain to the genus Halorubrum. The results of DNA-DNA hybridizations and physiological and biochemical tests allowed genotypic and phenotypic differentiation of strain AJ201 T from closely related species. Therefore, strain AJ201 T represents a novel species of the genus Halorubrum, for which the name Halorubrum arcis sp. nov. is proposed. The type strain is strain AJ201 T (=CGMCC 1.5343 T =JCM 13916 T ). (McGenity & Grant, 1995;Kamekura & Dyall-Smith, 1995) are widely distributed in hypersaline habitats (Grant & Larsen, 2001). At the time of writing, there are 16 species with validly published names in the genus Halorubrum, including three alkaliphilic species, Halorubrum vacuolatum (Mwatha & Grant, 1993; Kamekura et al., 1997), Hrr. tibetense (Fan et al., 2004) and Hrr. alkaliphilum (Feng et al., 2005), and 13 neutrophilic species, Halorubrum saccharovorum (the type species; Tomlinson & Hochstein, 1976), Hrr. sodomense (Oren, 1983, Hrr. lacusprofundi (Franzmann et al., 1988), Hrr. coriense (Nuttall & Dyall-Smith, 1993), Hrr. trapanicum (McGenity & Grant, 1995; Trüper, 2003), Hrr. distributum (Zvyagintseva & Tarasov, 1987;Oren & Ventosa, 1996) (Cui et al., 2006). Here, we report the taxonomic characterization of a neutrophilic strain belonging to the genus Halorubrum. Members of the genus HalorubrumStrain AJ201 T was isolated from mud of Ayakekum salt lake (37u 379 N 89u 299 E; 3884 m altitude) located in the Altun Mountain, in the northern part of the Qinghai-Tibet Plateau. The lake environment was described previously in detail (Xu et al., 2005a). The sample was incubated with S-G medium (Sehgal & Gibbons, 1960) for approximately 1 week and the liquid was plated by using a tenfold dilution series method. After 1-2 weeks of incubation at 37 u C, a representative red colony was picked and maintained on S-G medium. The strain was purified by repeated restreaking; purity was confirmed by the uniformity of cell morphology. The optimal conditions for growth were determined in S-G medium modified with 0.85-5.1 M NaCl or 0-1.0 M Mg 2+ .Phenotypic tests and lipid analysis were performed according to Xu et al. (2005b) as mentioned previously in the minimal standards for the description of new taxa in the order Halobacteriales (Oren et al., 1997). Detailed results of cell morphology, physiological tests and antibiotic sensitivity are given in the species description. A motile, pleomorphic, red-pigmented archaeon, strain EJ-52T, was isolated from water from Lake Ejinor, a saline lake in Inner Mongolia, China. Analysis of the almost-complete 16S rRNA gene sequence showed that the isolate was phylogenetically related to species of the genus Halorubrum, being most closely related to Halorubrum saccharovorum ATCC 29252T (96.1 % sequence similarity), Halorubrum lacusprofundi JCM 8891T (95.9 %), Halorubrum tibetense AS 1.3239T (95.2 %), Halorubrum alcaliphilum AS 1.3528T (95.2 %) and Halorubrum vacuolatum JCM 9060T (95.1 %). The polar lipids of strain EJ-52T were C20C20 derivatives of phosphatidylglycerol phosphate and phosphatidylglycerol phosphate methyl ester and a sulfated diglycosyl diether. Strain EJ-52T requires at least 2.5 M NaCl for growth and grows optimally at 3.4 M NaCl. The strain grows at 25–50 °C, with optimal growth occurring at 35–45 °C. Mg2+ is not required. The DNA G+C content is 64.2 mol%. On the basis of the data obtained in this study, strain EJ52T represents a novel species, for which the name Halorubrum orientale sp. nov. is proposed. The type strain is EJ-52T (=CECT 7145T=JCM 13889T=CGMCC 1.6295T).
