Grapefruit-derived nanovectors deliver miR-18a for treatment of liver metastasis of colon cancer by induction of M1 macrophages

Abstract: Liver metastasis accounts for many of the cancer deaths in patients. Effective treatment for metastatic liver tumors is not available. Here, we provide evidence for the role of miR-18a in the induction of liver M1 (F4/80+interferon gamma (IFNγ)+IL-12+) macrophages. We found that miR-18a encapsulated in grapefruit-derived nanovector (GNV) mediated inhibition of liver metastasis that is dependent upon the induction of M1 (F4/80+IFNγ+IL-12+) macrophages; depletion of macrophages eliminated its anti-metastasis eff… Show more

“…There are an increasing number of studies that demonstrate that the dysregulation of miRNAs is widely involved in the pathogenesis of colorectal cancer (1,9,11,12). However, the potential role and underlying mechanisms of miR-630 in colorectal cancer remain to be well demonstrated.…”

“…Altered expression of miRNAs contributes to the pathogenesis of the majority of types of human cancer, including colorectal cancer (9,10). To date, a variety of miRNAs, including miR-18a, miR-1, miR-133b and miR-374a, have been reported as regulators in a range of mechanisms in colorectal cancer (1,11,12). Therefore, dysregulation of miRNAs in colorectal cancer provides important opportunities for the development of future miRNA-based therapies (9).…”

“…Free GNVs and GNVs loaded with Dox (GNV-Dox), Dox+LA1 (GNV-Dox-LA1), and Dox+LA1+Psi (GNVDox-Psi-LA1) were prepared as previously reported [16]. Pre-prepared GNV-Dox was incubated with PEI for 60 min at room temperature; after centrifugation at 36, 000 rpm for 1 h, PEI-modified GNV-Dox (GNV-Dox-PEI) were resuspended and incubated overnight at 4°C with LA1 without or with Psi.…”

“…Kupffer cells are the resident macrophages of the liver and contribute to the metastatic process. (25,26) To elucidate the role of hepatic macrophages in liver metastasis, we determined the numbers of hepatic macrophages by immunofluorescence analysis. As shown in Figure 3(a), F4/80 + cells markedly accumulated into the metastatic tumors in WT compared with AT1aKO.…”
