Glycerol degradation in the thermoacidophilic crenarchaeonSulfolobus acidocaldariusinvolves an unusual glycerol-3-phosphate dehydrogenase

Christian Schmerling,
Carsten Schroeder,
Xiaoxiao Zhou
et al.

Abstract: Glycerol is highly abundant in nature and serve as carbon source for many organisms. Also, several Archaea have the genetic capacity to grow on glycerol but its degradation has so far only been studiedHaloferax volcanii.Herein, the thermoacidophilic crenarchaeonSulfolobus acidocaldariuswas shown to grow with glycerol as sole carbon and energy source. After uptake likely involving facilitated diffusion, glycerol is degraded via phosphorylation to glycerol-3-phosphate followed by oxidation to dihydroxyacetone ph… Show more

“…S. acidocaldarius strains MW001 (uracil auxotrophic) (31) and MW001 adapted to growth on glycerol designated herein MW00G (32) were cultured aerobically (Innova®44, New Brunswick, Germany) in Brock´s basal medium at 75°C, pH 3, 180 rpm (33) supplemented with 10 µg/ml of uracil as well as 0.1% (w/v) N-Z-amine and 0.3% (w/v) D-xylose (for induction of homologous overexpression). The Gelrite solidified Brock medium used for S. acidocaldarius MW001 and MW00G based cloning and transformation procedures was prepared as mentioned before (31).…”
