Glycan Microarrays for Decoding the Glycome

Abstract: In the last decade glycan microarrays have revolutionized the analysis of the specificity of glycan binding proteins, providing information that simultaneously illuminates the biology mediated by them and decodes the information content of the glycome. Numerous methods have emerged for arraying glycans in a ‘chip’ format, and glycan libraries have been assembled that address the diversity of the human glycome. Such arrays have been successfully used for analysis of glycan binding proteins that mediate mammalia… Show more

“…Microarray-Glycan microarrays are a unique tool for rapidly identifying possible receptors for pathogen attachment (58,59). A variety of viral proteins or whole viruses, including several RV VP8*s, have been analyzed on glycan arrays, which provided important information to direct biological and structural studies.…”

“…Glycan microarrays (20,21), which contain hundreds of carbohydrates immobilized on microscope slides, provide a multiplex assay for analyzing serum antiglycan antibodies that uses only minute amounts of valuable glycans and clinical samples (e.g., only 2-4 μL of serum). This approach, referred to as "glycoimmunomics," generates a comprehensive profile of antiglycan immune responses.…”

“…Arrays differ in the range of glycans/lectins and the manner in which they are displayed on the chip surfaces as this can be an influencing factor in the recognition of the glycan [21]. In many cases, specific thermodynamically stable conformation is needed for receptor recognition and high binding affinities are only achieved in the presence of specific neighbouring residues.…”
