GLS2 is transcriptionally regulated by p73 and contributes to neuronal differentiation

Abstract: The amino acid Glutamine is converted into Glutamate by a deamidation reaction catalyzed by the enzyme Glutaminase (GLS). Two isoforms of this enzyme have been described, and the GLS2 isoform is regulated by the tumor suppressor gene p53. Here, we show that the p53 family member TAp73 also drives the expression of GLS2. Specifically, we demonstrate that TAp73 regulates GLS2 during retinoic acid-induced terminal neuronal differentiation of neuroblastoma cells, and overexpression or inhibition of GLS2 modulates … Show more

“…However, GLS2 seems to support the growth and promote radiation resistance in some cancer types 61 . Indeed, GLS2 is induced by the tumor suppressor p53 and related proteins p63 and p73 55,56,62,63 , suggesting perhaps that it functions in resistance to radiation, or is important in cancers that still possess wild-type p53. Additionally, GLS2 is a critical downstream target of the N-MYC oncogene in neuroblastoma 64,65 .…”

“…Metabolic regulation by p53 family appears a key function for their cancer-related but also ageing-related phenotype. All the three members promote mitochondrial respiration thought regulation of different metabolic enzymes, including the glutaminase 2 (Gls2) (Adamovich et al, 2014;Amelio et al, 2014b;Bellomaria et al, 2010Bellomaria et al, , 2012Giacobbe et al, 2013;He et al, 2013;Hu et al, 2010;Sharif et al, 2015;Simon et al, 2014;Velletri et al, 2013;Viticchie et al, 2015). Gls2 regulates glutamine utilization, supplying anaplerotic flux of substrates resulting in promotion of mitochondrial activity and ATP synthesis (Amelio et al, 2014a;Maniam et al, 2015).…”
