Heterosis has been widely used in rice breeding, especially in improving rice yield. But it has rarely been studied in rice abiotic stress, including the drought tolerance, which is becoming one of the most important threaten in decreasing rice yield. Therefore, it is essential to studying the mechanism underlying heterosis in improving drought tolerance of rice breeding. In this study, Dexiang074B (074B) and Dexiang074A (074A) served as maintainer lines and sterile lines. Mianhui146 (R146), Chenghui727 (R727), LuhuiH103 (RH103), Dehui8258 (R8258), Huazhen (HZ), Dehui938 (R938), Dehui4923 (R4923), and R1391 served as restorer lines. The progeny were Dexiangyou (D146), Deyou4727 (D4727), Dexiang 4103 (D4103), Deyou8258 (D8258), Deyou Huazhen (DH), Deyou 4938 (D4938), Deyou 4923 (D4923), and Deyou 1391 (D1391). The restorer line and hybrid offspring were subjected to drought stress at the flowering stage. The results showed that Fv/Fm values were abnormal and oxidoreductase activity and MDA content were increased. However, the performance of hybrid progeny was significantly better than their respective restorer lines. Although the yield of hybrid progeny and restorer lines decreased simultaneously, the yield in hybrid offspring is significantly lower than the respective restorer line. Total soluble sugar content was consistent with the yield result, so we found that 074A can enhance drought tolerance in hybrid rice.

Quality-related traits are some of the most important traits in rice, and screening and breeding rice lines with excellent quality are common ways for breeders to improve the quality of rice. In this study, we used 151 recombinant inbred lines (RILs) obtained by crossing the northern cultivated japonica rice variety ShenNong265 (SN265) with the southern indica rice variety LuHui99 (LH99) and simplified 18 common rice quality-related traits into 8 independent principal components (PCs) by principal component analysis (PCA). These PCs included peak and hot paste viscosity, chalky grain percentage and chalkiness degree, brown and milled rice recovery, width length rate, cooked taste score, head rice recovery, milled rice width, and cooked comprehensive score factors. Based on the weight ratio of each PC score, the RILs were classified into five types from excellent to poor, and five excellent lines were identified. Compared with SN265, these 5 lines showed better performance regarding the chalky grain percentage and chalkiness degree factor. Moreover, we performed QTL localization on the RIL population and identified 94 QTLs for quality-related traits that formed 6 QTL clusters. In future research, by combining these QTL mapping results, we will be using backcrossing to aggregate excellent traits and achieve quality improvement of SN265.

KASP mapping of QTLs for yield components using a RIL population in Basmati rice (Oryza sativa L.)

Super Basmati is an elite variety with extra-long grains and superior quality but its yield is severely affected by water stress. Recombinant inbred lines (RILs) developed from the cross of Super Basmati and IR55419-04 (a coarse grained variety with high yield potential) were used to identify and map yield-related quantitative trait loci (QTLs) under normal field conditions. Genotypes for 244 KASP markers were obtained from 188 F9 RILs and a linkage map constructed of 1369.4 cM, with average marker interval of 10.5 cM. Eleven agronomic traits were phenotyped in RILs and parents. Composite interval mapping and inclusive composite interval mapping model identified 21 common QTLs related to nine agronomic traits; of the QTLs identified, twelve mapped at novel positions. LOD scores ranged from 2.51 for number of filled grains (qFG1.1) to 16.8 for plant height (qPH1.1). Four major effect QTLs could explain 20% of variation related for plant height, hundred-grain weight and grain width. Alleles from IR55419-04 improved grain filling, grain weight and grain width, while alleles from Super Basmati contributed to longer grains, panicles and flag leaves. Putative candidate genes were identified for 18 yield-related QTLs. These results validate the use of KASP genotyping for QTL mapping of yield-related traits in a bi-parental segregating population. SNPs in the QTLs identified in this study can be used in marker assisted selection for yield components to improve Basmati rice.
