Genetic rearrangements in the rfb regions of Vibrio cholerae O1 and O139.

Abstract: The recent emergence of a pathogenic new non-01 serotype (0139) of Vibrio cholerae has led to numerous studies in an attempt to identify the origins of this new strain. Our studies indicate .that 0139 strains have clear differences in the surface polysaccharides when compared with 01 strains: the lipopolysaccharide can be described as semirough. Southern hybridization with the 01 rjb region demonstrates that 0139 strains no longer contain any of the rjb genes required for the synthesis of the 01 0-antigen or i… Show more

“…A deletion in O1 El Tor C31 compared to other O1 V. cholerae in a region now identified as the MGC array has been reported in earlier studies (Barker & Clark, 1994;van Dongen & DeGraaf, 1986). Earlier studies have concluded that, unlike O139 Bengal strains, Arg3 is not derived from a V. cholerae O1 parent and has evolved independently (Stroeher et al, 1995(Stroeher et al, , 1997. This is supported by the recA phylogenetic analysis (Stine et al, 2000;Fig.…”

“…This finding is based first on the emergence of serogroup O139 (Bengal) in Bangladesh (CT positive) and Argentina (CT negative), each of which clearly evolved independently 33 . Second, the isolates from the sixth and seventh pandemic, and U.S. Gulf Coast belong to three different clones, each independently, evolved from environmental non-O1 V. cholerae isolates 16 .…”

“…et al., 1991, 1995aStroeher et al, 1992Stroeher et al, , 1995a) and a defective insertion-sequence-like element, IS1358d1, has been identified (Stroeher et al, 1995a). Interestingly, this element was also found linked to the rfb operon of the recently emerged V. cholerae serogroup O139 (Stroeher et al, 1995a).…”

“…Interestingly, this element was also found linked to the rfb operon of the recently emerged V. cholerae serogroup O139 (Stroeher et al, 1995a). IS1358 contains a putative transposase (tnpA) and has all the features of a transposable element (Stroeher et al, 1995a).…”
