Genetic linkage maps of 'Pêra' sweet orange and 'Cravo' mandarin with RAPD markers

Abstract: -The objective of this work was to construct linkage maps of 'Pêra' sweet orange [Citrus sinensis (L.) Osbeck] and 'Cravo' mandarin (Citrus reticulata Blanco) using RAPD markers and the pseudo-testcross strategy. The parents were chosen according to the resistance/susceptibility to citrus variegate chlorosis (CVC). The segregation of 176 markers was analyzed in 94 progeny of F 1 hybrids, which were obtained from controlled crossings. The linkage map of 'Pêra' sweet orange had 117 markers defined by 12 linkage … Show more

“…However, the number of mapped markers was insufficient to establish the nine linkage groups corresponding to the nine chromosomes present in haploid citrus. Some recent studies also focused on the genetic mapping of Citrus varieties [17,20,21,33]. The map of Gulsen et al [21] was the first C. clementina map, while Bernet et al [17] mapped Chandler pummelo and Fortune mandarin, a C. clementina × C. tangerina hybrid.…”

“…The hybrids were obtained by controlled crossing, with 'Cravo' mandarin as the female parent. Data from the segregation of 123 RAPD markers from 'Pêra' sweet orange and 53 from 'Cravo' mandarin, all obtained by Oliveira et al (2004), were used for genetic analyses. These markers were heterozygous for one of the parents (Aa) and recessive homozygous (aa) for the other, based on the presence of the RAPD bands.…”

“…Once the maps of each variety were constructed, the number, size and composition of the linkage groups, the length of the maps, the linear order and the distances among the markers were compared to the results obtained by Oliveira et al (2004). These authors used all markers to construct maps of Citrus, regardless of the absence or presence of skewed segregation.…”
