Abstract: Although ganglioside GD3 levels are highly elevated in malignant melanomas, the role of GD3 in melanomas' malignant properties has not been clearly shown. To investigate this problem, we genetically generated GD3-positive (GD3 ؉ ) transfectant cells from a GD3-negative (GD3 ؊ ) mutant line SK-MEL-28-N1 and analyzed the phenotypic changes in the transfected cells. GD3 ؉ cells showed markedly increased cell growth and invasive characteristics. Two bands that underwent stronger tyrosine phosphorylation in GD3 ؉ c… Show more

“…Tumor-associated ganglioside GD3 is known to be localized in GEM/rafts and generates signals by modulating receptor functions, leading to activation of downstream signaling pathways (35,36). For example, in human melanoma cells, expression of GD3 up-regulates phosphorylation levels of Akt, p130Cas, FAK, and paxillin after stimulation with fetal calf serum (37). GD3 also enhances adhesion signals by recruiting integrins to GEM/rafts (38).…”

“…55,56 BCAR1 signaling has also been associated with cell proliferation, survival, and migration/invasion in many tumor types besides breast cancer, including ovarian and prostate cancer, glioblastoma, melanoma, and hematopoietic malignancies. 11,12,17,27,[57][58][59][60][61][62][63] Accordingly, a great abundance of BCAR1 tyrosine-and serine/threonine-phosphorylated peptides have been detected in a wide variety of cancers (phosphosite.org) (Fig. 1).…”

“…Recent studies suggest that gangliosides are widely expressed on tumor cells (36) and disialyl gangliosides enhance tumor phenotype with multiple modalities (37). These reports demonstrate that GD3 promotes cell growth and invasion through phosphorylation of p130Cas, paxillin, and focal adhesion kinase (FAK) in melanoma cells (38). Furthermore, the Src family kinase Yes was found to link with p130Cas or FAK as an activated form to promote malignant properties of human melanoma cells expressing GD3 (37,39).…”
