Abstract: The rates of synthesis of histone H1 subtypes in synchronized mouse NIH 3T3 fibroblasts were compared with those of rat C6 glioma cells during the G0, G1, and S phases by using a combination of HPLC techniques and conventional gel electrophoresis. In the mouse cell line, all H1 subtypes, H1a-H1e including histone H1(0), were detectable. In the rat cell line, however, no histone H1a was found. H1c and H1e from both cell lines show in the quiescent state a relatively high specific activity comparable with that o… Show more

“…In mouse liver we found H1a, H1b, H1c, and H1 0 as pure subtypes, whereas one subfraction was a mixture containing the variants H1d and H1e (Fig. 1A) (28). Histone subtype H1t was obtained by means of reversed-phase HPLC fractionation of mouse testis (Fig.…”

“…The supernatant was mixed with 5 volumes of chilled acidified acetone and allowed to stand for 12 h at -20 °C. The precipitate was washed once with acetone, resuspended in water containing 10 mM 2-mercaptoethanol, and freeze-dried (Talasz et al, 1993).…”

“…16,17 Several reports have described a differential timing of the synthesis of the replication-dependent H1 histone subtypes. These studies were either focused on protein [18][19][20] or on mRNA levels, 21,22 and the results are in part contradictory. The present study is the first report about the cell cycle-dependent expression of all five replication-dependent H1 subtypes, the replacement histone H1.0 and the ubiquitous subtype H1x, both on mRNA as well as on protein level.…”
