Functional characterization of the Aspergillus fumigatus CRZ1 homologue, CrzA

Abstract: SummaryThe protein phosphatase calcineurin is an important mediator connecting calcium-dependent signalling to various cellular responses in multiple organisms. In fungi calcineurin acts largely through regulating Crz1p-like transcription factors. Here we characterize an Aspergillus fumigatus CRZ1 homologue, CrzA and demonstrate its mediation of cellular tolerance to increased concentrations of calcium and manganese. In addition to acute sensitivitiy to these ions, and decreased conidiation, the crzA null muta… Show more

“…52,53 All the comparisons with the deletion strains were performed with the CEA17 pyrG C . The media used were: complete medium composed for 2% w/v glucose, 0.5% w/v yeast extract, 2% w/v agar, trace elements (YAG) or YUU [YAG supplemented with 1.2 g (each) of uracil and uridine], and liquid YG or YG C UU medium with the same composition (but without agar).…”

“…Orthologues of Crz1 were recently shown to act as virulence factors in the human pathogen A. fumigatus (Cramer et al, 2008;Soriani et al, 2008) and the plant pathogen Botrytis cinerea (Schumacher et al, 2008). The fungal species surveyed in this study all contain a single orthologue of Crz1, except for S. pombe and R. oryzae which have two orthologues.…”

“…For instance, CrzA, the transcription factor downstream of calcineurin, plays an important role in germination and hyphal growth. A ⌬crzA mutant strain displays decreased conidiation, delayed and reduced germination rates, as well as defects in hyphal morphology and polarized hyphal growth (123,546). This mutant also has highly reduced virulence in a mouse model of invasive aspergillosis (123,546).…”
