Abstract: Nuclear magnetic resonance and visible spectroscopies were used to determine the thermodynamic parameters of the four hemes in cytochrome c3 from Desulfovibrio gigas at 298 and 277 K and to investigate the mechanism of electron/proton energy transduction. Data obtained in the pH range from 5 to 9 were analyzed according to a model in which the hemes interact with each other (redox cooperativities) and with an ionizable center (redox-Bohr cooperativities). The results obtained at the two temperatures allow the … Show more

“…The computer program used in this work fits the five center thermodynamic model, referred to above, to the NMR and visible data sets simultaneously (40). The half-height widths of the NMR signals were used as a measure of the uncertainty of each chemical shift.…”

“…Indeed, all the cytochromes c 3 studied to date showed a highly conserved heme core architecture (ref 46 and references therein) despite the low homology in their amino acid sequence. Nevertheless, the thermodynamic properties of D. Vulgaris, D. gigas, and D. desulfuricans 27774 (Dd27) cytochromes c 3 are all quite different (11,37,40,47).…”

“…This alternative mech-anism is achieved by the concerted effect of two positive redoxBohr effects governing a proton-assisted coordinated twoelectron transfer. Furthermore, these positive redox-Bohr effects are also responsible for the proton thrusting activity of this cytochrome (17).…”
