Abstract: We report the production of an important human therapeutic antibody in plant cell suspension cultures and the functional analysis of that antibody, including a comparison with the same antibody produced in CHO cells. We established transgenic tobacco BY2 suspension cell cultures expressing the human monoclonal antibody 2F5, which shows broadly neutralizing activity against HIV-1. The antibody was directed to the endoplasmic reticulum of the plant cells and was isolated by cell disruption, followed by protein A… Show more

“…All transformation constructs were based on the binary vector pTRA, a derivative of pPAM (GenBank accession no. AY027531) containing two tobacco RB7 scaffold attachment regions flanking the expression cassette (47). The coding regions of the 2G12 heavy and light chains (obtained from Polymun) contained an N-terminal signal sequence targeting the polypeptide to the secretory pathway.…”

“…To transiently transform tobacco BY2 suspension cultures (Nagata et al, 1992), they were mixed with transgenic agrobacteria in the presence of 200 mM acetosyringon (Sigma-Aldrich; Sack et al, 2007 To obtain stably transformed BY2 cultures, they were treated with cefotaxim (500 mg/mL; Duchefa) to eliminate agrobacteria and plated on MSBY agar containing kanamycin (100 mg/mL; Duchefa). After an incubation period of 3 to 4 weeks in the dark at 24°C, the appearing calli were transferred to fresh agar plates.…”

“…12,13 Additional antibody candidates for mucosal application are due to be tested in clinical studies in the near future, for example a plant-derived anti-HIV antibody envisaged as a vaginal microbicide. [14][15][16] The principal advantages of plant systems are rarely disputed, but the pioneers must succeed in their niche markets before plants become a mainstream platform and begin to compete on equal terms with mammalian cells. In many ways they are equivalent-plants can fold and assemble complex protein in the ©2 0 1 1 L a n d e s B i o s c i e n c e .…”
