This study aimed to evaluate the cause-effect relationship between canal preparation with ProTaper Universal (PTU) system and dentinal defects formation using micro-computed tomography (micro-CT) analysis. Forty mesial canals of mandibular molars with a type II Vertucci's canal configuration were scanned at an isotropic resolution of 14.16 µm. The sample was assigned to an experimental (n = 30) and a control (n = 10) groups, and the mesial canals were prepared with PTU system up to F2 instrument. The specimens from the experimental group were scanned and the cross-section images of the mesial roots, before and after preparation, were screened to identify the presence of dentinal defects. In the control group, the specimens were sectioned perpendicularly to the long axis of the root into 1-mm-thick slices (n = 80) and examined under optical microscope. Once a dentinal defect was detected, the slice was scanned through micro-CT. In the experimental group, dentinal micro-cracks were observed in 4,828 slices (24.04%). In all cross-section images, dentinal defects identified in the postoperative images were already present in the corresponding preoperative image. In the control group, 13 out of 80 slices (16.25%) had at least one dentinal defect visualized under stereomicroscopy, which was identified after a further micro-CT scanning. Micro-CT showed reliability as similar as optical microscopy in detecting dentinal defects, adding the possibility of tracking the dentinal tissue, before and after canal preparation, and providing a clear visualization of micro-cracks. Root canal preparation with PTU system did not induce the formation of new dentinal defects.

Aim: The aim of this study was to evaluate the fracture strength (FS) of bovine incisors with simulated incomplete rhizogenesis (IR) after different intraradicular treatments. Materials and methods:Incomplete rhizogenesis was simulated by sectioning the crowns and roots of 40 bovine incisors. Root canal preparation was performed and the samples divided into 4 groups (n = 10): GI-negative control with intra-radicular preparation; GII-positive control without intra-radicular preparation; GIII-glass fiber post cemented with resin cement and GIVroot canal obturation with Epiphany/Resilon system. In GIII and GIV, 4.0 mm apical plugs of mineral trioxide aggregate (MTA) were done. The samples were embedded into cylinders with polystyrene resin, and the periodontal ligament was simulated with a medium-viscosity polyether-based impression material (Impregum Soft). The specimens were submitted to compressive fracture strength test (0.5 mm/min at 135º in relation to the long axis of the tooth) in a mechanical testing machine MTS 810. Data were submitted to ANOVA and Tukey's test (α = 0.05).Results: GIII produced the highest FS values (p < 0.05). GI and GIV showed similar FS results (p > 0.05) but smaller than GII (p < 0.05). Conclusion:Glass fiber intraradicular posts provided the FS increase of teeth with simulated incomplete rhizogenesis. Epiphany/Resilon root canal filling with MTA plug did not provide the increase of the fracture strength to teeth with simulated incomplete rhizogenesis. Clinical significance:The fiber post reinforces the root structure of teeth with incomplete rhizogenesis.

The aims of this study were to evaluate the quality of canal filling using mineral trioxide aggregate (MTA) and gutta-percha (GP) in root canals, and to investigate the differences in the percentage of voids between mesial and distal canals by using microcomputed tomography. Twenty-two extracted human mandibular molars were instrumented using rotary files. Two teeth without canal fillings served as negative controls, and the other teeth were obturated with either ProRoot ® MTA or gutta-percha and AH Plus ® sealer. Obturated teeth were scanned with microcomputed tomography, and the percentage of voids (V %) was calculated. These specimens were also examined under a scanning electron microscope. The MTA group showed a significantly higher V % than the GP groups (p<0.05). The mesial canal groups showed significantly higher V% than did the distal canal groups (p<0.05). In orthograde filling, MTA exhibited significantly lower filling quality than did GP.
