Objective: The objective of the selected study was to design and formulate simvastatin-loaded pharmacosomes and then incorporate into a transdermal patch by solvent evaporation technique to enhance the solubility, bioavailability, and half-life of simvastatin. Methods: Simvastatin comes under the BCS-II class, which has low solubility and high permeability. Simvastain loaded pharmacosomes of six different formulations were prepared by taking simvastatin and soya lecithin in varying ratios and dissolved in a high polarity solvent dichloromethane and then subjected to the solvent evaporation method. Results: Formulated simvastatin-loaded pharmacosomes (SLP) were subjected to evaluation; out of six formulations, optimized formulation (F3) shown in vitro drug release of 86.88%; particle size of 151.6 nm with zeta potential of-16.5mV, which indicates good stability. SEM studies confirmed their smooth, porous structure with a number of nano-channels. The FT-IR spectra and DSC showed a stable character of simvastatin in a mixture of lipid and solvent shows compatible and revealed the absence of drug polymer interactions. The SLP was loaded into a transdermal patch by solvent evaporation method and evaluated for physical characteristics and results were found to be patch surface pH 6.15±0.08, thickness 0.146±0.0096 mm, weight uniformity 1.12±1.73, % swell-ability 13.50±0.028 for best patch formulation (F3). Conclusion: This research paper gives an outline on the significance of simvastatin-loaded Pharmacosomes as a transdermal patch for enhancing trans-permeation through the skin and its characterization and results. Through obtained results, it is concluded that pharmacosomes is a promising carrier to enhance the permeation of the selected drug through skin.

Lantana camara Linn is one of the plants that can help the wound healing process because it contains phenolic, flavonoids, and alkaloids that have antibacterial abilities against Staphylococcus epidermidis. but its use is still very simple, therefore it is necessary to develop traditional medicinal preparations in the transdermal patches with a combination of EC (Ethyl Cellulose) and PVP (Poly Vinyl Pyrolidone) polymers. This study aims to see the effect of using ethyl cellulose and polyvinyl pyrrolidone based on the physical characteristics of the Formulation. Lantana camara Linn leaf ethanol extract was obtained by maceration method using 96% ethanol solvent. The transdermal patch formulation was made with 7 formulations with a combination of EC: PVP based on a simple lattice design. Evaluation of the physical properties of the transdermal patch consisted of patch weight test, thickness test, folding endurance test, loss on drying, and moisture uptake. It can be concluded that the results of the evaluation of the preparation showed that F1 was the best preparation with polymer variations EC: PVP: PEG (200 mg: 250 mg: 50 mg) with a patch weight of 1.2213 ± 0.040; patch thickness 6.76 ± 0.251; folding endurance 310.33 ± 6.80; loss on drying 2.37 % ±0.36 and moisture absorption 3.68 % ±0.68.

Transdermal route of drug administration is novel as well as reliable means of sustained drug delivery. With more and more research being carried out in this field and increasing interest of researchers in this form of drug delivery, number of transdermal devices reaching the marketplace are expected to increase sharply. The aim of this review is to present latest explorations carried out in recent years utilizing possible drug candidates. Also, new polymer candidates along with novel penetration enhancers have been presented. For the purpose of study, various databases viz. Sciencedirect, Web of Knowledge, Pubmed, Google Scholar, etc. have been explored. After going through all the research work done by researchers in recent past it is appropriate to state that transdermal route is no longer reliant on few polymers and penetration enhancers but studies have now provided lot many more options for transdermal device formulation as evident from the data compilations. It can be concluded that most of the researchers have been utilizing HPMC as the preferred film forming polymer but recently use of Eudragit grades also has gained interest amongst scientists.
