Facile Synthesis of Fluorescent Hyper-Cross-Linked β-Cyclodextrin-Carbon Quantum Dot Hybrid Nanosponges for Tumor Theranostic Application with Enhanced Antitumor Efficacy

Abstract: Fluorescent hyper-cross-linked β-cyclodextrin-carbon quantum dot (β-CD-CQD) hybrid nanosponges of about 200 nm with excellent biocompatibility and strong bright blue fluorescence excited at 365 nm with a high photoluminescence quantum yield (PLQY) of 38.0% were synthesized for tumor theranostic application by facile condensation polymerization of carbon quantum dots (CQDs) with β-cyclodextrin (β-CD) at a feeding ratio of 1:5. The DOX@β-CD-CQD theranostic nanomedicine, around 300 nm with DOX-loading capacity of… Show more

“…The nanocarriers equipped with defined functionality for tailored interaction through 'host-guest chemistry' , and biodegradable properties can ferry poorly soluble agents to selectively target cancer cells with stem-like properties and controllably release their payload in response to the environment, thus resulting in an improved antitumor activity in vivo. Pei et al [133] linked the β-cyclodextrin (β-CD) on the CDs to form β-CD-CDs, which further DOX can be loaded via host-guest complexation. The DOX -loading capacity can reach 39.5% and release in the weakly acidic tumor microenvironment.…”

“…Besides, protonation of AUR’s ether bonds and amide bond cleavage under acidic conditions may also be a cause of AUR release from the nanogel. 34 – 36 Taken together, the release of AUR and CDDP were obviously affected by the acidic environment, and the behavior of differential release in physiological conditions and tumor environment helped to improve the selectivity of drugs.…”

“…CQDS have been widely used in biological imaging. Carboxyl-functionalized carbon quantum dots are known to exhibit high quantum yield and abundant surface carboxyl groups, which could be used as cross-linking agents for β-CD, endowing fluorescent functions to β-CD (Figure 8 a) 112 . This cross-linked polymer has been shown exhibit both fluorescence imaging properties and drug delivery functions.…”
