The aim of this study was conducted to investigate a method to production purify IgY and compare the capacity between Taiwan native Tsaiya ducks (layer type) and Leghorn laying hens. Ten Taiwan native Tsaiya ducks and 10 white Leghorn laying hens (20 wk-old) were injected with 250 μL of complete Freund's adjuvant containing bovine serum albumin (BSA, 200mg) during week 1 of the trial. The boost injection, during week 2 and 4 of the trial was comprised 250 μL of incomplete Freund's adjuvant containing BSA was conducted to promote IgY production. Eggs were collected for IgY purification from the week 5 of the trial. Yolk protein was first extracted using isopropanol or triton and followed by salting out with ammonium sulfate. After dialysis, gel filtration chromatography and affinity chromatography were used to purify the IgY antibody. Yolk proteins extracted by isopropanol or triton from duck eggs were 43% and 13%, and 61% and 16% from hen eggs, respectively. After salting out, the duck IgY had a purity of 53−57% with a titer of 26.2−29.9 unit/mg protein, while the hen IgY had a purity of 45−50% with 24−29.5 unit/mg protein. After purification by gel filtration chromatography, the purity of the duck IgY was increased to 89−90% with 44.24−50.62 folds purification and 415−451 unit/mg protein. The purity of the chicken hen IgY was increased to 85−87% with 29.85−30.38 folds purification and 240−277.6 unit/mg protein. After further purification by affinity chromatography, duck IgY purity was increased to 91−92% with 45−56.26 folds purification and 459−461 unit/mg protein; chicken hen IgY purity was increased up to 93−95% with 33.58−59.49 folds purification and 312−470 unit/mg protein. This procedure is promising for purification and production of the IgY antibody from Taiwan native Tsaiya ducks and/or white Leghorn laying hens, and both of them are good producer of IgY.

Urease inhibition with oral antibodies is a promising strategy in the treatment of Helicobacter pylori infection. This study aimed to investigate the advantages of the producing method of anti-urease IgY and its inhibitory effect on H. pylori infection on experimental animal. We started the process of producing IgY with the preparation of H. Pylori's urease. Then the eggs from the hens that were immunized with the prepared urease were collected to be used for IgY extraction. We divided 18 Swiss albino mice into 2 groups to test the inhibitory effect of the IgY. One group was fed the food containing IgY and the other was not. In the end, the mice were examined to determine the reactivity of anti-H. pylori urease IgY in the sera and the inflammation and infection status of the stomach biopsies. The ELISA results confirmed the presence of IgY in the hens' sera. The IgY from egg yolks was purified using ammonium sulfate precipitation. Pathological examination of 55.6% of the stomachs from the infected mice from the study group showed considerable reduction of inflammation in the tissue compared to those from the control group (p < 0.0001), and 22.2% of the mice from the study group was free from gastritis, based on the histopathology results. Our method of IgY purification can be promisingly applied in large-scale production. Oral administration of urease-specific IgY is successful in inhibiting H. pylori colonization in the inocculated mice and can be considered an alternative to antimicrobial therapy.

The use of immunoglobulin is successfully applied in different areas of research, diagnostics, medical application and biotechnology. Egg yolk immunoglobulin (IgY) can successfully compete with immunoglobulin (IgG) produced in the blood of mammals. Recently, successful progresses have been achieved in Japan through industrialization of IgY technology. Using IgY has been shown to provide a safer, more efficient and less expensive method for managing disease-causing pathogens. Helicobacter pylori (H. pylori), a spiral Gram-negative microaerophilic pathogen, it infects over 50% of the population worldwide, and is recognized as the etiologic agent of gastritis, peptic ulcer, and has been linked to the development of gastric adenocarcinoma and mucosa associated lymphoid tissue lymphoma. It is found that urease is the most abundant protein of H. pylori. Urease is recognized as an essential factor in the organism colonization of the gastric mucosa. The eradication of H. pylori by administration of oral antimicrobials is not always successful and may be associated with adverse effects. Therefore, several treatment regimens have emerged to cure H. pylori infection. Accordingly, a novel approach in prevention and reduction of H. pylori infection has been reported based on production of urease-specific immunoglobulin that can suppress the bacterial colonization through urease-binding by anti-H. pylori urease IgY (IgY-urease). The use of IgY against a pathogenic factor of H. pylori will be a prudent way to suppress the infection.
