Expression of interleukin‐1α and interleukin‐1 receptor type I genes in murine brown adipose tissue

Abstract: At thermoneutral conditions, high steady-state levels of transcripts for both IL-lct and its receptor IL-1RtI were found in specialized thermogenic organ, brown adipose tissue (BAT) of adult mice, as compared with the levels in lymph nodes, brain and spleen. A pronounced decrease of IL-1 c~ mRNA level in BAT was observed after lipopolysaccharide (LPS) administration and after exposure to cold. Likewise, LPS decreased the IL-I RtI mRNA level and depressed also the expression of cold-inducible genes for the BAT-… Show more

“…The role of adipose tissues in immune response and inflammation has been suggested by previous studies (Choy et al, 1992; White et al, 1992; Burysek et al, 1993; Cianflone and Maslowska, 1995; Peake et al, 1997; Pond, 1999) including our recent demonstration that preadipocytes present macrophage‐like functions (Cousin et al, 1999). The present work is the first evidence for a modification of these functions in preadipocytes under inflammatory stimuli such as thioglycollate treatment and/or pathological situation such as obesity.…”

“…Cultured cells were washed with PBS prior to total RNA isolation according to the guanidinium/phenol/chloroform procedure [12]. Northern blots (20 μg total RNA aliquots) were sequentially hybridized with cDNA probes radiolabeled with [α‐ 32 P]dCTP (3000 Ci/mmol) using the Random Priming Kit (Boehringer Mannheim) essentially as before [13]. Mouse IL‐1α cDNA was 1.6‐kbp fragment cut by Pst I and Eco RI from the pIL1301 plasmid provided by P.T.…”

“…Mouse IL‐6 cDNA (1.1‐kbp Eco RI fragment) was kindly provided by Van Snick (Ludwig Institute for Cancer Research, Brussels, Belgium) [15]. The probe for β‐actin was identical with that used earlier [13]. Radioactivity on the membranes was quantified by PhosphorImager analysis system (Molecular Dynamics).…”

“…The intracellular pathway involved is unknown, but it is probable that the response is mediated by the IL‐1R1 receptor that is found in the tissue. The summary is based on data by Burysek et al 65–67…”
