Expression of hPOT1 in HeLa Cells and the Probability of Gene Variation of hpot1 Exon14 in Endometrial Cancer are Much Higher than in Other Cancers

Abstract: To investigate the expression of hPOT1 in the HeLa cell line and screen point mutations of hpot1 in different tumor tissues a two step osmotic method was used to extract nuclear proteins. EMSA was performed to determine the expression of hPOT1 in the HeLa cell line. PCR was also employed to amplify the exon14 sequence of the hpot1 gene in various of cancer tissues. A SV gel and PCR clean-up system was performed to enrich PCR products. DNAStar was used to analyse the exon14 sequence of the hpot1 gene. hPOT1 was… Show more

“…POT1 expression was higher in the biopsy tissue of cervical cancer patients than that in normal cervical smears. In previous studies, POT1 had high and strong expression in HeLa cells (12). Consistent with this, POT1 also had high expression in the biopsy tissue of cervical cancer patients, as well the HeLa cell line.…”

“…Consistent with this, POT1 also had high expression in the biopsy tissue of cervical cancer patients, as well the HeLa cell line. Expression of POT1 has been identified in some cancer cell lines such as gastric cell lines SGC-7901 and MKN28 and larynx squamous carcinoma cell lines Hep-2 and Hep-2R (12).…”

“…The protein and/or mRNA levels of the most conserved out of all shelterin proteins, POT1 (26) were increased in many different cancers including gastric, thyroid, breast (199, 278, 279) and in endometrial cancers (280). Higher levels of point mutations in the POT1 gene were observed in endometrial cancers, revealing that genetic variations in POT1 may lead to carcinogenesis in the endometrium (280). Simultaneous conditional inactivation of the shelterin protein POT1a with the tumor suppressor p53 in endometrial epithelial cells in a murine model, induced type II metastatic adenocarcinomas in 100% of the animals by 15 months (281).…”

“…Thus, we also selected shelterin proteins TERF1/2 for examination. The third shelterin subunit, POT1 was not included as it has already been examined in the endometrium [ 47 ]. TERF2IP was included in our study as it binds to TERF2, therefore is structurally relevant [ 48 ].…”
