Expression of Antiviral Activity and Induction of 2′,5′-Oligoadenylate Synthetase by Conceptus Secretory Proteins Enriched in Bovine Trophoblast Protein-11

Abstract: Establishment of pregnancy in cattle has been proposed to depend on production of a conceptus protein, bovine trophoblast protein-1 (bTP-1), which has a high degree of sequence homology with bovine interferon-alpha (bIFN-alpha), especially the alpha II subfamily. A preparation of bovine conceptus secretory proteins enriched for bTP-1 has antiviral and physico-chemical properties similar to other bIFN-alpha. Antiviral activity is initially detectable in uterine flushings on Day 14 of pregnancy, when the concept… Show more

“…Conceptus length and antiviral activity in the uterus (a measure of IFNT2 bioactivity; Alexenko et al1997) were highly variable, as is typical at this stage of pregnancy (Short et al 1991, Mann & Lamming 2001, Block et al 2007), but the largest conceptuses were in the CSF2 group. Antiviral activity tended to be greater for cows receiving CSF2 conceptuses and IFNT2 gene expression was 22 times greater for CSF2 conceptuses.…”

“…The OAS1 expression was amplified during days 15-18 in heifers 36 and cow 37 . Shirasuna et al (2012) documented that, the OAS1 expression did not vary in pregnant and nonpregnant cows 25 .…”

“…Testing this hypothesis revealed that pregnancy and IFNT induced the production of uncharacterised proteins by the endometrium [17]. Some of these proteins and genes induced by pregnancy/IFNT were later revealed to be classical Type I IFN stimulated genes (ISGs) 2',5'-oligoadenylate synthetase 1, 40/46 kDa; OAS1 [18,19] With the widespread availability of large-scale transcriptomic analysis, the molecular mechanisms required to establish uterine receptivity to implantation and pregnancy recognition are beginning to be elucidated. The initial in vivo analysis of the effect of the conceptus on endometrial gene expression was carried out using subtracted cDNA libraries [28] and a model of monozygotic twins to reduce genetic variation.…”
