Abstract: BackgroundCannabinoid receptor 1 (CB1) is expressed in certain types of malignancies. An analysis of CB1 expression and function in Hodgkin lymphoma (HL), one of the most frequent lymphomas, was not performed to date.Design and MethodsWe examined the distribution of CB1 protein in primary cases of HL. Using lymphoma derived cell lines, the role of CB1 signaling on cell survival was investigated.ResultsA predominant expression of CB1 was found in Hodgkin-Reed-Sternberg cells in a vast majority of classical HL c… Show more

“…The over expression of cannabinoid receptors and elevated endocannabinoid levels have also been reported in a variety of different cancer types such as prostate, skin, hepatocellular carcinoma, colon, endometrial sarcoma, glioblastoma multiforme (GBM), meningioma and pituitary adenoma (see table 1) Xu et al, 2006;Pisanti et al, 2013). CB 1 receptors are also up-regulated in Hodgkin lymphoma cells and also in chemically induced hepatocarcinoma (Benz et al, 2013;Mukhopadhyay et al, 2015). Also the expression of CB 1 and CB 2 receptors was found to increase in mantel cell lymphoma and fatty acid amide hydrolase (FAAH) expression was reduced when compared to non-malignant B-cells (Islam et al, 2003;Ek et al, 2002;Wasik et al, 2014).…”

“…qRT-PCR experiments were performed according to the protocol in Benz et al (2013). 46 The reaction volumes contained 17 ml as follows: 10 ml PCR-MasterMix (Promega Inc., M7505), 0.5 ml of left and right primer (25 pM), 0.25 ml of Eva Green dye (Biotium, 31000), 4.75 ml of nuclease free water (Promega Inc., P1193) and 1 ml of cDNA.…”

The PCR reaction was performed in a rotor-cycler (Rotor-Gene TM RG 6000; Corbett Research, Pty Ltd) and consisted of 5 steps: initial denaturation at 95 C, followed by 40 cycles of denaturation at 94 C (3 s), annealing at 64 C (30 s ; followed by a stepwise decrease of 1 C per cycle during the first 5 cycles down to an annealing temperature of 59 C), elongation at 72 C (30 s) and fluorescence detection at 80 C (15 s).…”
