Cerebellar Purkinje cells (PCs), the sole output neurons in the cerebellar cortex, play an important role in the cerebellar circuit. PCs appear to be rather sensitive to aging, exhibiting significant changes in both morphology and function during senescence. This article reviews such changes during the normal aging process, including a decrease in the quantity of cells, atrophy in the soma, retraction in the dendritic arborizations, degeneration in the subcellular organelles, a decline in synapse density, disorder in the neurotransmitter system, and alterations in electrophysiological properties. Although these deteriorative changes occur during aging, compensatory mechanisms exist to counteract the impairments in the aging PCs. The possible neural mechanisms underlying these changes and potential preventive treatments are discussed.

The herbicide 2,4-dichlorophenoxyacetic acid (2,4-D) is widely used in several countries. Research on the neurotoxicity of 2,4-D has been focused on the central nervous system, and little is known about its effects on the myenteric plexus. Therefore, to elucidate the neurotoxicity of 2,4-D and the viability of its use, we investigated the effects of daily intake of 5 mg 2,4-D/kg for 60 days on the myenteric plexus neurons of the rat ileum using quantitative and morphometric analyses. Twenty male Wistar rats aged 60 days were divided into two groups (n=10 rats/group). The group E animals received daily 5 mg doses of 2,4-D/kg diluted in 1 mL of water, whereas the group C animals were not treated with the herbicide. The animals were euthanized with anesthetic after 60 days; subsequently, the ileum was dissected out, and procedures were performed to visualize the total population of neurons (Giemsa staining), the nitrergic neurons (NADPHd+), and the estimated population of cholinergic neurons (NADPHd-). The results were statistically analyzed and compared between groups. In group E, the densities of Giemsa-stained neurons and NADPHd- neurons decreased (p<0.05) by 7% and 10%, respectively, whereas the density of NADPHd+ neurons remained constant. The cell body area was 5.8% greater (p<0.05) for the NADPHd+ neurons but remained unchanged for the neurons stained with Giemsa. These results suggest that 2,4-D causes a reduction in neuronal density, particularly for cholinergic neurons, and promotes an increase in the cell body area of nitrergic neurons, leading to hypertrophy.

ΣΚΟΠΟΣ ΚΑΙ ΜΕΘΟΔΟΙ: Η οδός μηνύματος του νεοπλασματικού αυξητικού παράγοντα TGF-β έχει περιγραφτεί ως ογκοκατασταλτικό αλλά και ως ογκοπροαγωγό μονοπάτι στην βιολογία των κυττάρων. Επιπρόσθετα, η απενεργοποίηση μορίων προσκόλλησης της κυτταρικής μεμβράνης όπως η E-Cadherin, αλλά και πολυλειτουργικών πρωτεϊνών όπως η p21, η οποία αναστέλλει τις κυκλοεξαρτόμενες κινάσες, παίζουν σημαντικό ρόλο στο μεταστατικό δυναμικό του κολοορθικού καρκίνου. Ο σκοπός της παρούσας μελέτης είναι να αξιολογηθεί η έκφραση των πρωτεϊνών του σηματοδοτικού μονοπατιού TGF-β, της E-Cadherin και της p21, και τα αποτελέσματα αυτά να συσχετιστούν με κλασσικούς προγνωστικούς παραμέτρους, όπως το στάδιο της νόσου, τις λεμφαδενικές μεταστάσεις και την επιβίωση των ασθενών. Η έκφραση των παραπάνω πρωτεϊνών αξιολογήθηκαν με ανοσοϊστοχημική μέθοδο σε 195 δείγματα από ασθενείς με καρκίνο του παχέος εντέρου. ΑΠΟΤΕΛΕΣΜΑΤΑ: Οι παράγοντες TGF-β, p21 και E-Cadherin είχαν θετική έκφραση σε (71,28%), (65,13%) και (50,26%) αντίστοιχα στα δείγματα ασθενών με καρκίνο του παχέος εντέρου. Η συσχέτιση της έκφρασης των παραγόντων αυτών με τα κλινοπαθολογικά χαρακτηριστικά των δειγμάτων, έδειξε ότι η θετική έκφραση του TGF-β και η χαμηλή έκφραση των E-cadherin και p21 είχαν στατιστικά σημαντική αντιστοιχία με το στάδιο κατά TNM της νόσου (P=0,0012), (P=0,0363) και (P=0,0452) αντίστοιχα. Η θετική έκφραση του παράγοντα TGF-β, σχετιζόταν επίσης με την παρουσία λεμφαδενικών μεταστάσεων (P=0,0089). Οι καμπύλες επιβίωσης κατά Kaplan-Meier έδειξαν επίσης, μια σαφή συσχέτιση της συνολικής επιβίωσης των ασθενών με κολοορθικό καρκίνο, με την υψηλή έκφραση TGF-β καθώς και τη χαμηλή έκφραση p21 και E-Cadherin. ΣΥΜΠΕΡΑΣΜΑΤΑ: Τα αποτελέσματα της παρούσας μελέτης δείχνουν ότι οι παράγοντες του μονοπατιού TGF-β, η πρωτεΐνη p21 και η πρωτεΐνη E-Cadherin έχουν στενή συσχέτιση με το στάδιο του καρκίνου του παχέος εντέρου και αποτελούν πολύτιμους δείκτες προγνώσεως.
