Evidence that a second tumor antigen coded by adenovirus early gene region E1a is required for efficient cell transformation.

Abstract: The expression of the adenovirus (Ad) early coding region la (Ela) is required for virus-induced cell transformation and for the activation of other viral early genes and some cellular genes. Two overlapping early mRNAs of 13S and 12S that are transcribed from this region code for a 289-amino acid protein and a 243-amino acid protein, respectively. Earli Oncogenic transformation by adenoviruses is a function of both the Ela and Elb (mp 4.5-11.3) regions as determined by the analysis of virus-transformed cells … Show more

“…Replication defective adenoviral vectors are deleted of sequences spanning EIA, EIB, and a portion of the E3 region, impairing the ability of this virus to replicate or transform nonpermissive cells ( 13). In the vector AdCMVlacZ, the early enhancer/promoter of the cytomegalovirus (CMV)' was used to drive transcription of lacZ with an SV40 polyadenylation sequence cloned downstream from this reporter.…”

“…1). The presence of conserved domain 3 in the ElA products appears to correlate strongly with the ability of the ElA gene products to trans activate the other virus genes efficiently, while various transformation-related activities of the ElA region appear to be independent of this domain (8,15,21,23,26,42,43,50,63,73).…”
