Reports over the years have demonstrated toxic side effect-including reproductive toxicity-of tamoxifen (TAM), a drug of choice in the management of primary breast cancer. Chlorogenic acid (CGA), a dietary polyphenol, reportedly elicits beneficial pharmacological effects. However, the impact of CGA on TAM-associated reproductive toxicity is absent in the literature. We, therefore, experimented on CGA's effect and TAM-mediated reproductive toxicity in rats. Cohorts of rats were treated with TAM (50 mg/kg) or co-treated with CGA (25 or 50 mg/kg) for 14 consecutive days. The result showed that treatment of CGA significantly increases testosterone, LH, and FSH levels compared to the TAM group. However, prolactin level was markedly decreased after pretreatment of CGA in TAM-treated rats. CGA abated TAM-induced decreases acid phosphatase, alkaline phosphatase, and antioxidant enzymes in the testis. CGA alleviated TAM-facilitated surges of reactive oxygen and nitrogen species, myeloperoxidase, nitric oxide, interleukin-1β, and tumor necrosis factor-alpha in rats epididymis and testes. Additionally, CGA increased anti-inflammatory cytokine-interleukin-10-, suppressed caspase-3 activity, and reduced pathological lesions in the examined organs of rats co-treated with CGA and TAM. CGA phytoprotective effect improved reproductive function occasioned by TAM-mediated toxicities in rats, by abating oxido-inflammatory damages and downregulating apoptotic responses. Practical applications CGA protects against the damaging oxido-inflammatory responses incumbent on TAM metabolism. As an antioxidant abundant in plant-derived foods, CGA reportedly protects against inflammatory damage, hypertension, and neurodegenerative diseases. We present evidence that CGA ameliorates TAM-induced reproductive dysfunction by suppressing oxidative and inflammation stress downregulate apoptosis and improve reproductive function biomarker in rats.

The Effects of Costus Root Aqueous Extract against Schistosomiasis, Liver Damage and Oxidants in Mouse Models of Schistosoma mansoni Infection

Schistosomiasis is one of the major human parasitic diseases in many developing countries that causing chronic ill health in humans. The present work aimed to study the possible curative role of Costus (Saussurea lappa) root extract against Schistosoma mansoni infection induced liver toxicity, cytokines, oxidative stress and DNA damage in mice. 40 adult male micewere divided into four groups (G1, control; G2, Costus; G3, infection with S. mansoni live cercariae; G4, S. mansoni infection followed by Costus treatment). By comparison to control, mice with S. mansoni infection exhibited significantly (p<0.01) higher levels of plasma TNF-α, faecal and hepatic eggs load, hepatic DNA damage and MDA, as well as significantly (p<0.01) lower levels of hepatic GSH, catalase and SOD. Meanwhile, by comparison to mice with S. mansoni infection, infected mice treated with Costus root extract had significantly (p<0.01) lower levels of faecal and hepatic eggs load, granuloma diameter, TNF-α in plasma, hepatic MDA, DNA damage, as well as significantly higher levels of hepatic GSH, catalase and SOD. The conclusion derived was that free radicals are formed by the host-parasite interaction because the parasite induces alterations in the hepatic antioxidants of the host to be able to scavenge the radicals as oxidative stress makes it difficult for the parasite to overpower the host's immune response.

Interest in green synthesis methods for nanoparticle production with potential use in biology, pharmacy, and medicine is a forefront field in research. Among the routes described in the literature, the use of plant extracts as reducing and capping agents is one of the most promising. Turnera diffusa commonly called Damiana is a wild shrub traditionally used as aphrodisiac, diuretic, and for cancer and diabetes treatments. Its extracts are rich in flavonoid, phenolic, terpenoid, and other active compounds that possess antiallergic, antiviral, and antifungal activities. Besides, flavonoids have been reported to have a redox potential sufficient for metal ion reduction. Thus, this study reports a novel and facile method for green gold nanoparticle (AuNPs) synthesis using T. diffusa extracts. The samples were fully characterized by scanning electron microscopy, acrylonitrile butadiene styrene ultraviolet-visible, dynamic light scattering, X-ray photoelectron, and inductively coupled plasma optical emission spectroscopies. The results showed high efficiency of T. diffusa extracts to reduce and stabilize the AuNPs which showed spherical shape, 24 nm diameter, negative surface charge, and high colloidal stability at biological pH and alkaline media. Additionally, its antimicrobial activity was evaluated showing high antiseptic capacity against gram-negative Salmonella enteritis and Escherichia coli ATCC 8739 bacteria.
