Abstract: Ellagitannins constitute the largest group of hydrolyzable tannins of plants, and, from this group, casuarictin (Casu) was identified in some plant species. However, to our knowledge, no investigation of secretory phospholipase A2 (sPLA2) inhibition by Casu has been performed yet. Casuarictin was isolated by chromatography n-butanol (n-BuOH) partition of Laguncularia racemosa leaves. The pharmacological and biological effects of Casu were evaluated on isolated sPLA2 from the rattlesnake (Crotalus durissus terr… Show more

“…Tannins are common in plant extracts investigated for anti-venom activity, and various studies have examined the effects of purified tannins on snake venoms [41][42][43][44][45]61,62] and their toxins, especially PLA 2 [15,63]. Kupussamy and Das [61] investigated the protective effect of tannic acid injected subcutaneously (s.c.) on the lethality and other actions of Crotalus admanteus (eastern diamonback rattlesnake) venom in mice.…”

“…The protocols used here did not allow a clear distinction between these possibilities. In this regard, Rodrigues et al [15] have shown that casuarictin, an ellagitannin isolated from the white mangrove (Laguncularia racemosa) forms a 1:1 molar complex with C. d. terrificus PLA 2 without precipitating out of solution; this solubility meant that the effect of this compound could be studied on a variety of biological activities mediated by the PLA 2 . Since these authors did not study intact crotoxin (PLA 2 + crotapotin, such as occurs in the venom), it is unclear whether interaction with this ellagitannin would be different (both in molar ratio and solubility).…”

“…Hydrolyzable tannins occur naturally in fruits, such as strawberries, raspberries, blackberries, and pomegranates. Ellagitanins and gallotannins are heavily present in seeds, pericarp, and peel of pomegranate (Rodrigues et al, 2019;Kandylis and Kokkinomagoulos, 2020). Punicalagin belongs to the ellagitannins group and gives the pomegranate skin its yellow color.…”

“…Secretory A2 phospholipases (sPLA2) from snake venom in species such as Crotalus durissus terrificus (Cdt) have structural and functional similarity to most human sPLA2, and both have common enzymatic functions and properties [1]. These sPLA2 enzymes have been used as research tools both in silico and experimentally in the search for new anti-inflammatory compounds.…”
