Abstract: Background Delayed hypersensitivity (HS) reactions to potentiated sulfonamide antimicrobials occur in both dogs and humans, and involve an intermediate hydroxylamine metabolite that is detoxified by cytochrome b5 and NADH cytochrome b5 reductase. Hypothesis/Objectives We hypothesized that polymorphisms in the genes (CYB5A and CYB5R3) encoding these 2 enzymes would be associated with risk of sulfonamide HS in dogs. Animals A total of 18 dogs with delayed HS to potentiated sulfonamide antimicrobials and 16 d… Show more

“…The start codon in exon 1M was found to be 337 bp downstream of the 5′ end of the gap, whereas the end of exon 1 was located 173 bp upstream of the 3′ end of the gap. The information about polymorphism in the canine CYB5R3 gene was compared with the previous studies . In our study, a new SNP in the promoter region, c.1‐234 T/G, was identified, and all the tested dogs were homozygous for the G allele at this position.…”

“…A limitation of our study was our inability to evaluate expression levels of CYB5R3 mRNAs and b5R proteins in erythrocytes from dogs. Nonsynonymous SNPs have been reported to be able to affect not only mRNA stability, and thus protein expression, but also enzymatic activity . We tried to analyze the b5R protein concentrations in dog erythrocytes by immunoblotting using an anti‐human b5R antibody, but no b5R protein signals were detected.…”

“…In a population of dogs expanded from our original report, (Funk‐Keenan et al., ) the CYB5R3 729G allele remained overrepresented in HS dogs (0.792) compared to dogs tolerant to sulfonamide antibiotics (0.550); thus, the G allele may indeed predispose to sulfonamide HS. However, it is important to note that 30% of tolerant dogs are still homozygous for 729GG.…”

“…CYB5R3 729A>G allele and genotype frequencies were also compared between dogs hypersensitive and tolerant to sulfonamide antibiotics (24 HS, 20 TOL). Of these, 34 were previously reported (Funk‐Keenan et al., ) and 10 were newly recruited dogs. There was no significant difference in age ( p = .39) or sex ( p = .69) between groups (Table ).…”
