Evaluation of API Coryne in comparison with conventional methods for identifying coryneform bacteria

Abstract: A study was performed to evaluate a new manual miniaturized system, API Coryne (API-bioMerieux, Inc., La Balme les Grottes, France), in which conventional biochemical methods were used to identify 240 isolates of coryneform and related bacteria. A total of 40% of the isolates were excluded from the study because they could not be identified by conventional methods. Identifications of the 240 isolates obtained with API Coryne showed a 97.6% concordance with conventional methods (79% after 24 h of incubation, 21… Show more

“…A variety of biochemical and serological tests have been used for identification of A.pyogenes. These methods included conventional methods (1 1, 12, 13, 19) and commercial identification systems (7,12,14). However, the latter were performed with heterologous test systems (14) or only with a small number of A.pyogenes-isolates (7).…”

“…The first commercially available system specifically designed for identification of members of the genus Corynebacterium is the Rapid CORYNE by API-bioMerieux. This system was recently evaluated by Freney et al (5), who tested 240 strains of gram-positive bacilli belonging to eight genera, with the vast majority from the genus Corynebacte-* Corresponding author. t Present address: Department of Obstetrics and Gynecology, University of Washington, Seattle, WA 98195. rium or the CDC coryneform groups.…”

“…In addition, DNase activity was observed after 2 days, tyrosine hydrolysis also became positive after 2 days whereas hydrolysis of casein became positive after 10 days only, and starch hydrolysis was negative. When applying the commercial API Coryne system (bioMe èrieux) [10] we observed the numerical code 2472325 (a seven digit code corresponding to 21 biochemical reactions) for both strains.…”
