Evaluation of an Enzyme-Linked Immunosorbent Assay Based on Araraquara Virus Recombinant Nucleocapsid Protein

Abstract: Laboratory diagnosis of hantavirus cardiopulmonary syndrome (HCPS) in Brazil has been performed mostly by detection of IgM antibodies to recombinant antigen purified from Sin Nombre virus and Andes virus (ANDV). Recently, a recombinant nucleocapsid (rN) protein of Araraquara virus (ARAV), a Brazilian hantavirus, was obtained in Escherichia coli. To evaluate ARAV rN as antigen for antibody detection, serum samples from 30 patients from Argentina seropositive for hantavirus were tested. All samples were positive… Show more

“…The sera were tested with enzyme-linked immunosorbent assay (ELISA) for the presence of immunoglobulin M (IgM) and immunoglobulins G (IgG) antibodies to hantavirus, by using the recombinant nucleocapsid (N) protein of the Araraquara virus (ARAV), according to the description of Figueiredo et al (3) . Samples with an optical density equal to or greater than the average of the negative controls plus three standard deviations at a dilution of 1:400 were considered positives.…”

“…In this study, protocols of IgM-ELISA and IgG-ELISA were performed as previously described by Figueiredo et al, using the recombinant N protein of Araraquara hantavirus as antigen (ARAV rN) produced in E. coli 8 . Figueiredo et al, found that ELISA has high sensitivity (97.2%), specificity (100%), positive predictive value (100%), and negative predictive value (98.1%).…”
