ETV1 positively regulates transcription of tumor suppressor ARF

Abstract: ETV1 (ETS variant 1) is a transcription factor from the ETS family and an oncogene in several types of human malignancies. Paradoxically, a predicted inactivating mutation in ETV1 was previously found in a clone of HT1080 cells with reduced activity of p53. We report that elevated expression of ETV1 makes p53-null tumor cells hypersensitive to restoration of said tumor suppressor. Furthermore, elevated levels of either wild-type ETV1 or its truncated derivative, dETV1, which mimics the product of an oncogenic … Show more

“…Importantly, we present evidence that ETV1 suppresses motility and migration of melanoma cells and therefore function as a tumor suppressor. To the best of our knowledge, there is only one report, on fibrosarcoma, that suggests an inhibitory role for ETV1, resulting in increased expression of mRNA of p14ARF [ 52 ], a known tumor suppressor which activates p53 [ 53 ]. Worth noting, the majority of melanomas in fact lack ETV1 amplification, representing > 80% of the cases [ 44 ].…”

“…16 hours after plating, cells were transfected with miR-497 (1 pmol / well) or non-targeting siRNA (1 pmol / well), pRSV-βgal (constitutive β-galactosidase expression vector; 70 ng / well), and an appropriate luciferase reporter plasmid (30 ng / well) using Lipofectamine 2000 (Life Technologies) per manufacturer's protocol. Luciferase activity was measured and normalized for that of β-galactosidase as described before [ 57 ].…”

“…Prostate-specific antigen (PSA) expression is also enhanced by ETV1 (Gasi, 2014). We currently have a limited understanding of how molecular pathways are affected by elevated ETV1 levels in prostate cancer (Gasi, 2014; Zynda, 2013). …”
