Abstract: Our data suggest that LA stimulates neurite outgrowth through the activation of ERK signaling, an effect mediated through a ROS-dependent mechanism. This article is part of a Special Issue entitled: 11th European Symposium on Calcium.

“…These data add to previous work from our laboratory and others suggesting a critical role for this signaling pathway in neuroblast differentiation (12,49,(61)(62)(63)(64)(65). These studies urge caution in the clinical use of nonspecific tyrosine kinase inhibitors, which may inhibit this important differentiation pathway and lead to disease recurrence.…”

“…Recent evidence suggests that Erk signaling is also critical for retinoic acid-and α-lipoic acid-induced NB differentiation and cell-cycle exit via upregulation of p21 (31,32), suggesting activity of this differentiation pathway in response to multiple stimuli. Interestingly, of the signaling components involved in the differentiation pathway we have identified (including FGF2, FGFR1, TβRIII, Erk1/2, Id1, and p21), only TβRIII expression varies by stage of disease, while correlating with improved prognosis, highlighting its importance (Figure 1, Supplemental Figure 6, E and F; and negative data not shown).…”

“…FGF2 has been shown to promote neuronal differentiation of neural-crest tumor cells via the Erk MAPK pathway (26,(28)(29)(30). Erk signaling is also critical to retinoic acid-and α-lipoic acid-induced neuroblast differentiation (31,32), suggesting a broader involvement for this pathway in NB differentiation.…”

“…Mitogen-activated protein kinase (MAPK) and Akt signaling are crucial for neurite outgrowth and are activated by natural compounds, 31,32) and are associated with neural protective effects after drug-induced injury. 33) JNK mediates not only cell survival. 15) JNK is induced during neuronal differentiation in the developing brain in PC 12 cells, as well as in other neuronal cell lines.…”
