BackgroundStaphylococcus aureus is associated with chronic mastitis in cattle, and disease manifestation is usually refractory to antibiotic therapy. Biofilm production is a key element of S. aureus pathogenesis and may contribute to the treatment failure that is consistently reported by veterinarians. Minas Gerais State is the largest milk-producing state in Brazil, and the characterization of bacterial isolates is an important aspect of disease control for dairy farmers. Here, we investigated the potential of S. aureus isolated from bovine mastitis to produce slime and biofilm in a skim-milk medium and classified the isolates according to their agr type.ResultsSlime was detected using the Congo Red agar (CRA) test in 35.18% (19/54) of the strains; however, 87.04% (47/54) of the strains were considered biofilm-positive based on crystal violet staining. Compared to TSB supplemented with 0.25% glucose, skim milk significantly increased the production of biofilm, but this effect was only observed in slime-producing strains. The bacteria belonged to agr groups I (12/54), II (34/54), III (6/54), and IV (2/54), and bacteria in agr group III were found to be stronger biofilm producers than those in groups I and II. Again, milk had a significant influence only on slime-positive agr I and II isolates, revealing an association between milk and slime.ConclusionsThe present study demonstrated that skim-milk medium and slime production are two factors that together influence biofilm formation by bovine strains of S. aureus. A predominance of bacteria belonging to agr group II was observed, and bacteria from agr group III showed the highest proportion of biofilm producers. The majority of bacteria characterized in this study formed biofilm in milk, which suggests that biofilm formation has an important role in the virulence of S. aureus isolated from bovine intramammary infections.

Antimicrobial Activity of Copaifera Spp. Against Bacteria Isolated From Milk of Cows With Mastitis

The antimicrobial activities of the oleoresin (OR) and the essential oil (EO) of Copaifera spp. were checked against microorganisms isolated from milk samples of cows diagnosed with grade III subclinical mastitis. The OR had good antimicrobial activity (MIC ≤ 100 µg/mL) against samples of coagulase-positive Staphylococcus, coagulase-negative Staphylococcus, Streptococcus of groups C, F, and G, and Corynebacterium spp. Meanwhile, the EO had good antimicrobial activity (MIC≤100 µg/mL) against coagulase-negative Staphylococcus and Corynebacterium spp. The OR and the EO were inactive and weak to inactive, respectively, against Escherichia coli. Overall, the OR had better antimicrobial activity than the essential oil against the 55 bacterial isolates (p<0.0001). The GC/MS analysis identified sesquiterpenes in EO and by the ESI FT-ICR MS method, the identification of diterpenic acids in OR was possible. Therefore, this raw plant material is promising for the development of phytotherapeutic drugs against bovine mastitis.Keywords: bovine mastites; ESI FT-ICR MS; essential oil; minimum inhibitory concentration; oleoresin. A atividade antimicrobiana da oleorresina e do óleo essencial de Copaifera spp. foi verificada frente aos micro-organismos isolados de amostras de leite de vacas diagnosticadas com mastite subclínica grau III. A oleorresina de Copaifera ssp. apresentou boa atividade antimicrobiana (CIM≤100 µg/mL) frente às amostras de Staphylococcus coagulase positivo, Staphylococcus coagulase negativo, Streptococcus do grupo C, F, G e Corynebacterium spp. Já o óleo essencial de Copaifera spp. apresentou boa atividade antimicrobiana (CIM≤100 µg/mL) frente aos Staphylococcus coagulase negativo e Corynebacterium spp. Em relação à Escherichia coli, a atividade antimicrobiana da oleorresina e o óleo essencial foi inativa e fraca à inativa, respectivamente. Conclui-se que, frente às 55 bactérias isoladas, a olerresina apresentou melhor atividade antimicrobiana do que o óleo essencial de Copaifera spp. (p<0,0001). A análise por CG/EM identificou sesquiterpenos no óleo essencial e pelo método ESI FT-ICR MS, foi possível identificar ácidos diterpenos na oleorresina de Copaifera spp. Por fim, sugere-se que a oleorresina de Copaifera spp. é uma matéria-prima vegetal promissora no desenvolvimento de medicamento fitoterápico para o tratamento de mastite bovina.Palavras-chave: concentração inibitória mínima; ESI FT-ICR MS; mastite bovina; óleo essencial; oleorresina.

Enterotoxin-encoding genes in Staphylococcus aureus from buffalo milk

This paper investigated the occurrence of Staphylococcus aureus and the detection of enterotoxin-encoding genes of these strains in milk collected from 30 Murrah buffaloes used to produce dairy products in Brazil. A total of 68 strains of Staphylococcus aureus were found as identified by conventional laboratory tests, and thus screened for sea, seb, sec, sed, see, seg, seh and sei enterotoxin-encoding genes by polymerase chain reaction (PCR). Twelve strains containing enterotoxin-amplified genes were found, with higher expression for the sei and seh genes. These results can be attributed to animal health and inadequate cleaning of the equipment, indicating the need for better quality control in animal production and health lines. The results of this study with the presence of pathogens and their enterotoxigenic potential indicate a source of food poisoning, as well as being a pioneering study in the detection of new enterotoxins for buffalo milk.
