Abstract: Background: DNA hypomethylation at the F2RL3 locus has been associated with both smoking and atherosclerotic cardiovascular disease; whether these smoking-related associations form a pathway to disease is unknown. F2RL3 encodes protease-activated receptor 4, a potent thrombin receptor expressed on platelets. Given the role of thrombin in platelet activation and the role of thrombus formation in myocardial infarction, alterations to this biological pathway could be important for ischemic cardiovascu… Show more

“…A total of 1452 CpG sites were mentioned once or more in at least two of the publications, 441 CpG sites were mentioned once or more in at least three publications, and two CpG sites were mentioned in ≥ 6 publications (Table 2 ). The two latter CpG sites are cg01656216 near gene ZNF438 (mentioned in [ 10 , 22 , 30 , 60 , 65 , 108 ]) and cg03636183 near gene F2RL3 (mentioned in [ 49 , 69 , 75 , 79 , 113 ], Table 3 ). These two CpG sites are also part of the methylation clocks.…”

“…Three out of six papers mentioning cg01656216 [ 22 , 30 , 108 ] have vascular disease as an outcome, whereas the three other publications had investigated epigenetic age [ 10 , 60 , 65 ]. In the case of cg03636183, three out of six publications had coronary heart disease or myocardial infarction as the outcome [ 75 , 79 , 113 ], and the other three had investigated smoking or air pollution [ 12 , 49 , 69 ].…”

“…Platelet activation is enhanced in patients with myocardial infarction and is a hallmark of acute coronary syndrome (Frossard et al, 2004). Recent study established a direct link between smoking-induced hypomethylation of F2R like thrombin or trypsin receptor 3 (F2RL3) and platelet activity, providing further evidence for its contribution to smoking related cardiovascular risk (Corbin et al, 2022). Here, using the COmics webserver, we investigate the molecular network around the CpG cg05575921 at aryl hydrocarbon receptor repressor (AHRR) locus, a well-known smoking marker (Zaghlool et al, 2018), that is also significantly associated with arteriosclerosis, which is driven by inflammation, and carotid plaque (Ammous et al, 2022; Zhang et al, 2017).…”

“…El tabaquismo tiene efectos en la metilación del ADN en muestras de sangre completa (12)(13)(14), células bucales (15) y tejido pulmonar (16,17). Entre los loci identificados y que presentan metilación diferencial relacionada con el consumo de tabaco y la arteriosclerosis destacan CpG localizados en los genes F2RL3 (F2R like thrombin or trypsin receptor 3) y ALPPL2 (9,18).…”
