Abstract: Variation in DNA methylation (DNAm) is associated with lifestyle factors such as smoking and body mass index (BMI) but there has been little research exploring its ability to identify individuals with major depressive disorder (MDD). Using penalised regression on genome-wide CpG methylation, we tested whether DNAm risk scores (MRS), trained on 1223 MDD cases and 1824 controls, could discriminate between cases (n = 363) and controls (n = 1417) in an independent sample, comparing their predictive accuracy to pol… Show more

“…In a transcriptomic study of schizophrenia, two genes harbouring the 108 loci were differentially expressed in the prefrontal cortex 56 . In addition, epigenetic risk scores for depression are largely independent of the polygenic genetic risk scores 57,58 . Given these previous findings of weak relationships between differential expression, methylation, and genetic hits, our number of highlighted genes is not unexpected.…”

“…BeadChip (Illumina Inc., San Diego, California) in 9778 participants across two processing sets (set 1 N=5,190 (comprised related individuals), set 2 N=4,583 (comprised unrelated (to each other and to set 1) individuals)) and quality controls were carried out using standard methods outlined in supplementary methods as described previously (41,42). The final DNAm dataset comprised data for 5,087 individuals in set 1 and 4,450 individuals in set 2.…”

“…Health and lifestyle data were also collected, as were physical measurements such as height and weight, from which BMI was derived as a covariate of interest 32 . Smoking status was collected from GS baseline assessments as were the number of smoking pack years-full details have been reported by Barbu and colleagues 31 .…”

“…Since this measure of inflammation may be less prone to the acute effects of cross-sectional measures of serum CRP described above, DNAm CRP may provide a more stable signature of exposure to chronic inflammatory states compared to cross-sectional serum CRP 29,30 . Indeed, methylation risk scores of other phenotypes including MDD itself have proven discriminatory utility, with comparable/better performance compared to genetic risk scores 31 .…”
