Epidemiology and phospholipase activity of oral Candida spp. among patients with central nervous system diseases before and after dental cleaning procedure

Abstract: Patients suffering of diseases that affect central nervous system may be considered more susceptible to the infectious diseases of mouth. Sixty-nine patients suffering of cerebral palsy, Down’s syndrome and metal retardation were submitted to saliva examination for the presence of Candida spp. before and after a procedure of dental cleaning. The isolates were submitted to assay for verifying phospholipase production. 55.10% of the patients provided isolation of Candida spp. The frequency of isolation obtained … Show more

“…Briefly, 100 μL of serially diluted GQAs (13.87 to 222 μg/mL) was incubated with precultured Vero cells (6 × 10 4 cells/mL) in Dulbecco’s modified Eagle medium (DMEM) using 96-well culture plate. Following 48 h of incubation, the cytotoxic effects of GQAs on test cells were determined quantitatively using the MTT assay method as described by Vistica et al ( 44 ) and Ribeiro et al ( 45 ).…”

“…Phospholipase activity (P z ) was determined by measuring the ratio of colony diameter to the total diameter of both the colony and the precipitation zone. A P z value of <1 indicates enzymatic activity, while a value of 1 denotes no activity; i.e., the lower the value of P z , the higher the phospholipase activity ( 45 ).…”

“…For qualitative analysis of phospholipase production, 1 µl of inoculum was placed at the center of the SDA plate containing 1% peptone, 3% glucose, 5.73% NaCl, 0.055% CaCl 2 , and 10% of egg yolk emulsion in the absence and presence of morin (at MBIC), and the plates were then incubated at 37 • C for 72 h (Ribeiro et al, 2010). The diameter of the precipitation zone around the colony was determined.…”
