Enzyme‐Mediated Tumor Starvation and Phototherapy Enhance Mild‐Temperature Photothermal Therapy

Abstract: Compared with conventional tumor photothermal therapy (PTT), mildtemperature PTT brings less damage to normal tissues, but also tumor thermoresistance, introduced by the overexpressed heat shock protein (HSP). A high dose of HSP inhibitor during mild-temperature PTT might lead to toxic side effects. Glucose oxidase (GOx) consumes glucose, leading to adenosine triphosphate supply restriction and consequent HSP inhibition. Therefore, a combinational use of an HSP inhibitor and GOx not only enhances mildtemperatu… Show more

“…This is the principle behind the proposed cancer starvation therapy, which effectively starves the cells by depleting their glucose supply ( Kim and Dang, 2006 ; Li et al, 2018 ). This approach has stimulated much interest ( Fu et al, 2018 ; Yu et al, 2018 ; Xie et al, 2019 ) and the glucose oxidase GOx has attracted attention ( Fu et al, 2019 ; Yang et al, 2019 ; Gao et al, 2020 ; Ren et al, 2020 ) for its role in the oxidation of glucose to gluconic acid, a reaction that can be harnessed to restrict the glucose supply to tumor cells ( Jain, 2014 ; Gao et al, 2020 ).…”

“…Starvation therapy blocking the nutrient metabolism pathways deprives cancer cells of energy, which meanwhile restricts adenosine triphosphate supply to inhibit HSPs and replenishes H 2 O 2 for intensified PTT/CDT (G. Gao et al, 2020). For instance, a versatile antitumor copper molybdenum sulfide (Cu 2 MoS 4 ) was loaded with glucose oxidase (GOx) for cooperative PTT/CDT/immunotherapy/starvation therapy (Chang et al, 2019).…”

“…The inorganic PTAs include metal or metal-containing nanomaterials including gold nanostructures [ 11 , 12 ], palladium-based nanostructures [ 13 15 ], iron or copper-containing nanoparticles (NPs) [ 16 19 ], transition metal chalcogenides [ 20 ], and quantum dots [ 21 , 22 ]. On the other hand, organic materials [ 23 ] including near-infrared (NIR) dyes represented by cyanine dyes [ 24 32 ], conjugated polymers (e.g., polydopamine (PDA), polyaniline, polypyrrole, and poly(3,4-ethylenedioxythiophene)) [ 33 36 ], and some carbon-based nanomaterials (most of them are organic materials; as represented by graphite-related nanostructures) [ 37 41 ] have also been utilized for PTT.…”
