Chemiluminescence (CL) is a very promising method for applications in analytic chemistry and biological systems due to the elimination of the external light source, which could reduce autofluorescence interference and improve the signal‐to‐noise ratio (SNR) and tissue penetration depth. The fluorescent dyes, chemiluminescent initiator, and reaction temperature are three crucial factors that determine the chemiluminescence efficiency. Up to now, significant development of chemiluminescence probes has been reported for high‐performance imaging and sensing, including the systems based on luminol, imidazopyrazinone, peroxyoxalate, and Schaap's dioxetane. In this review, we give an overview of the current chemiluminescence probes and their bioimaging and biosensing applications. We also provide perspectives on designing chemiluminescent probes with good efficiency and longer emission wavelengths.

Chemiluminescence (CL) reactions have been widely employed and explored over the past 50 years because they offer unique light emission upon a defined chemical stimulus. In this Minireview, we focus on peroxyoxalate (PO) compounds because they feature very high quantum yields tuneable over the entire visible spectrum, allowing for visible‐light detection by the naked eye without the necessity for expensive analytical instruments. Although analytical methods have been extensively described, PO‐CL read‐out is a strongly emerging field with ample industrial potential. The state‐of‐the‐art PO‐CL detection read‐out systems for various key analytes is here explored. In particular, structural requirements, recent developments of PO‐CL read‐out probes and current limitations of selected examples are detailed. Furthermore, innovative approaches and synthetic routes to push the boundaries of PO‐CL reactions into biological systems are highlighted. Underpinned by recent contributions, we share perspectives on embedding PO‐CL molecules into polymeric materials, which they consider the next step in designing high performance solid‐phase read‐out systems.

The outbreak of African swine fever (ASF) has caused significant economic losses to animal husbandry worldwide. Currently, there is no effective vaccine or treatment available to control the disease, and therefore, efficient disease control is dependent on early detection and diagnosis of ASF virus (ASFV). In this study, a chemiluminescent immunoassay (CLIA) was developed using the ASFV protein p54 as a serum diagnostic antigen and an anti-p54 monoclonal antibody. After optimizing the working parameters of the CLIA, the sensitivity of the established CLIA was 1:128, ASFV-specific serum antibody was identified, and there was no cross-reaction with other swine virus antibodies. After testing 49 clinical serum samples, the consistency rate between the CLIA and the World Organization for Animal Health (OIE) recommended commercial kit was 100%. Thus, this CLIA had a high degree of specificity, sensitivity, and reliability, and could be used as a rapid detection method for epidemiological investigations of ASFV infection.
