Abstract: Background: MicroRNAs (miRNAs) negatively regulate gene expression at a post-transcriptional level. Dicer, a cytoplasmic RNase III enzyme, is required for the maturation of miRNAs from precursor miRNAs. Dicer, therefore, is a critical enzyme involved in the biogenesis and processing of miRNAs. Several biological processes are controlled by miRNAs, including the regulation of T cell development and function. T cells generate reactive oxygen species (ROS) with parallel H+ extrusion accomplished by the… Show more

“…Myricetin is a flavonoid with potentant antioxidant properties and considered to be an excellent scavenger of intracellular reactive oxygen species 57,58 . Studies have shown that oxidative stress, including increased accumulation of superoxide and hydroxyl radicals, induces cytosolic acidification in a wide range of tissue types 59–61 . In the presence of 1% DMSO or 10 μM myricetin, we observed a significant attenuation in extracellular acidification, similar to that observed with indole.…”

“…In VSMCs, luminal acidification may be involved in phosphate-induced calcification [46] and calcium-phosphate-induced cell death [47]. NHE1 may further be required for the generation of oxidative stress [48][49][50][51][52], which has in turn been shown to participate in the orchestration of osteogenic signaling [53][54][55][56][57]. In addition, NHE1 activity may be linked to production of inflammatory cytokines [58], which may contribute to osteogenic mechanisms in VSMCs [59].…”

“…20,000 cells/sample in the flow cytometer were acquired using the recommended software for data acquisition. Obtained values were corrected for autofluorescene of control cells without dye 66 , 67 . Geometric mean of the FL-1 signal intensity was used to show the amount of Fluo-4 and H2DCFDA fluorescence intensity (separate measurements).…”

“…The murine primers used to detect IL-1β, IL-2, IL4, IL-6, IL-10, IL-12, TNFα, IFNγ, TGF-β, and NOS2 expression are summarized in Table 1 . For amplification of different miRNAs, hsa-miR-9-5p LNA PCR primer set (#204513), hsa-miR-10a-5p LNA TM PCR primer set (#204778), has-miR-99b-5p LNA PCR primer set (#205983), hsa-miR-146a-5p LNA PCR primer set (#204688), mmu-miR-155-5p LNA PCR primer set (#205930), and reference 5S rRNA primer set (#203906) were used and the reaction was set up as recommended by Exiqon 67 , 69 .…”
