Enhanced production of osteopontin in multiple myeloma: clinical and pathogenic implications

Abstract: Summary. In this study, we examined osteopontin (OPN) production in myeloma cells and plasma OPN levels in multiple myeloma (MM) patients. We assessed OPN production in bone marrow cells (BMCs) by immunocytochemistry and enzyme-linked immunosorbent assay (ELISA). We also assessed OPN production in various B-cell malignant cell lines, including three myeloma cell lines by reverse transcription polymerase chain reaction (RT-PCR) and Western blotting. In addition, we measured plasma OPN concentrations by ELISA in… Show more

“…This finding agrees with some [25, 26], but not all, investigations [20, 27]. Furthermore, Saeki et al, Scudla et al, and Minarik et al have observed differences in plasma OPN levels between monoclonal gammopathy of undetermined significance and MM [26, 28–30].…”

“…The association between OPN and solid cancers has been reported in hepatocellular carcinoma, pancreatic Cancer and lung cancer [20,21,22]. Increased serum OPN concentrations occur in chronic myeloid leukemia, multiple myeloma, and acute myeloid leukemia [23]. Higher OPN were associated with significant shorter survival times in AML patients in a 1-year survival analysis [24].…”

“…In one study, OPN from bone marrow derived cells enhanced migration of MCF-7 breast cancer cells in a Transwell co-culture system [71]. Bone marrow cells in overt myeloma patients produce more OPN than myeloma precursor disease [72]. In addition, myeloma cells express and bind to OPN [72, 73].…”
